摘  要:目的探讨甲基转移酶抑制剂5-氮-2-脱氧胞苷(DAC)对胆管癌细胞TFK-1侵袭力的影响及其机制。方法采用Transwell法检测胆管癌细胞在不同浓度DAC(0、10、20、40、80、160μmol·L-1)作用下侵袭力的变化;Western-blot法检测DAC作用后Wif-1蛋白的表达。结果 DAC对胆管癌细胞侵袭力有显著性抑制(P<0.05),且呈浓度依赖性;Wif-1蛋白在TFK-1细胞中未见表达,DAC处理后Wif-1蛋白再现表达。结论 DAC对胆管癌TFK-1细胞的侵袭有抑制作用,此作用可能是由Wif-1基因去甲基化来实现。Objective To investigate the effect of DNA methyltransferase inhibitor 5-aza-2-de-oxycytidine (DAC)on the invasion of TFK-1cholangiocarcinoma cells and its mechanism of action.Methods Transwell method was used to detect the invasion of cholangiocarcinoma cells treated with different concentrations of DAC (0,10,20,40,80 and 160 μmol·L^-1 ).Western-blot method was used to detect the expression of Wif-1 protein.Results DAC inhibited the invasion of cholangiocarcinoma cells in a concentration-dependent manner.The expression of Wif-1 protein was not observed in cholangiocarcinoma cells,but was detected after treatment with DAC.Conclu-sion DAC can inhibit the invasion of cholangiocarcinoma cells,and the mechanism may be in-volved in the demethylation of Wif-1 gene.
