摘 要:目的疫苗佐剂研制是疫苗研制的关键环节之一。而中药多糖安全、价廉、来源广泛,研究报道有免疫增强作用。故本研究选用黄芪多糖(Astragalus polysaccharide,APS)、香菇多糖(Lentinan,LNT)、枸杞多糖(Lyeium barbarum polysa Ceharide,LBP)以不同浓度组成不同的中药配伍组群,遴选出最有应用前景、最有效的疫苗佐剂配伍。方法以APS、LNT、LBP为3因子,上述三种中药多糖的18mg/kg,36mg/kg,54mg/kg为3水平,交叉设计出9个不同剂量的佐剂配伍组群,对免疫抑制小鼠进行灌胃处理后,观察小鼠的脾脏指数、胸腺指数和脾脏淋巴细胞增殖的变化。结果 1与正常对照组比较,第1、2、3、6、7、9的中药多糖配伍组,能明显提高小鼠脾脏指数,差异有统计学意义(P<0.05);2与模型对照组比较,第1中药多糖配伍组,能明显提高小鼠脾脏指数,差异有统计学意义(P<0.05);3与正常对照组比较,第4、8中药多糖配伍组,能显著增加小鼠脾T淋巴细胞增殖活性,差异有统计学意义(P<0.05);4与模型对照组比较,第1、2、5、6、7、9的中药多糖配伍组,能显著增加小鼠脾T淋巴细胞增殖活性,差异有统计学意义(P<0.05)。结论综合上述结果显示在APS、LNT、LBP9个配伍组中,以第1组(即三种中药多糖各18mg/kg)为最佳配伍组,不仅对非特异性免疫功能有增强作用,对特异性免疫功能也具有增强作用。可成为最有效的免疫佐剂配伍。Objective Developing a vaccine adjuvant is one of the key aspects of vaccine development. The traditional Chinese medicine polysaccharide is safe,inexpensive,widely available,which studies have reported immune enhancement. There-fore,this study selected Astragalus polysaccharide(APS),Lentinan(LNT),LyeiumbarbarumpolysaCeharide(LBP)to compose dif-ferent traditional Chinese medicine compatibility groups with different concentrations and selected the most promising and most ef-fective vaccine adjuvant compatibility at last. Methods Taking APS,LNT,LBP as 3 factors,three kinds of concentrations,18mg/kg,36mg/kg,54mg/kg of the above three Chinese herbal as 3 levels and making a crossover design of 9 different doses of adjuvant compatibility group. After intragastric administration on the immunosuppressed mice,observing the spleen index,thymus index and the T lymphocyte proliferation changes in spleen. Results ①Compared with normal control group,the Chinese medicine polysac-charide compatibility group 1,2,3,6,7,9 can significantly improve the spleen index,the difference was statistically significant(P〈0. 05);② Compared with the model control group,the first compatibility group of Chinese medicine polysaccharide can significant-ly increase the spleen index,the difference was statistically significant(P〈0. 05);③Compared with the normal control group,the Chinese medicine polysaccharide compatibility group 4,8 can significantly increase the T lymphocyte proliferation activity in spleen of the mice,the difference was statistically significant(P〈0. 05);④Compared with the model control group,the Chinese medicine polysaccharide compatibility group 1,2,5,6,7,9 can significantly increase the splenic T lymphocyte proliferation,the difference was statistically significant(P〈0. 05). Conclusion Based on the results above showed that in the APS,LNT,LBP 9 compatibility group ,group 1(three Chinese polysaccharides each 18mg/kg)was the best compatibility group which not only has the enhancing
