摘  要:目的:通过免疫小鼠获得抗人结缔组织生长因子( CTGF )抗体的基因,构建抗人CTGF单链抗体( scFv)噬菌体展示文库,并通过生物淘选获得抗人CTGF的单链抗体。方法抗人CTGF单链抗体噬菌体展示文库经过4轮富集筛选之后,得到抗CTGF单链抗体,用ELISA及基因测序检测CTGF单链抗体亲和力。结果经过4轮富集筛选,得到了抗人CTGF scFv;ELISA检测显示抗人CTGF scFv与CTGF具有高亲和力,基因测序显示抗CTGF scFv基因全长767 bp,编码255个氨基酸,具有和人CTGF结合的特异性。结论本实验通过噬菌体展示技术筛选出抗人CTGF单链抗体,为以后CTGF单链抗体的功能研究奠定基础。Objective To construct single chain Fv( scFv )library of human connective tissue growth factor( CTGF)by assembling the gene from immunized mice to phagemid PAK100,and to obtain the anti human CTGF scFv by phage display technique. Methods Anti human CTGF ScFv was obtained from the scFv library after four rounds of bio-panning. The affinity was examined using ELISA assay and DNA sequence. Results High affinity anti human CTGF scFV was obtained. Sequence analysis of positive clones displayed the anti human CTGF scFv was 767 bp,coding 255 amino acids. Conclusions The yield from this study gives a reference for the investigation on the bioactivity of anti-human CTGF scFv.
