摘 要:为了探讨细胞外信号调节激酶(ERK)在阪崎肠杆菌侵袭脑微血管内皮细胞过程中的作用,用ERK抑制剂(PD98059)预处理体外培养的小鼠脑微血管内皮细胞,再通过细菌侵袭实验观察细胞侵袭力的变化,进而通过Western-blot实验检测ERK的活化情况,最后利用免疫荧光方法观察肌动蛋白微丝的重排情况。结果显示:PD98059可以降低阪崎肠杆菌侵袭进入脑微血管内皮细胞的数量、阻碍阪崎肠杆菌侵袭脑微血管内皮细胞引起的ERK磷酸化、抑制阪崎肠杆菌侵袭所导致的脑微血管内皮细胞细胞骨架的重排。因此,阪崎肠杆菌通过ERK的活化影响脑微血管内皮细胞的微丝重排,进而侵袭进入脑微血管内皮细胞。To investigate the role of the extracellular signal -regulated kinase (ERK) in Cronobacter sakazakii invading into brain microvascular endothelial cells, the mouse brain microvascular endothelial cells were treated with ERK inhibitor (PD98059). The bacterial invasive ability was performed by bacterial invasion assay; the activation of ERK was assessed by Western - blot ; the actin cytoskeleton rearrangements were visualized by immunofluorescence. The results showed that ERK inhibitor ( PD98059 ) decreased the invasion of Cronobacter sakazakii into brain microvascular endothelial cells, and the phosphorylation of ERK was increased in brain microvascular endothelial cells infected with Cronobacter sakazakii. The Cronobacter sakazakii - induced ERK phosphorylation and actin filaments rearrangements in brain microvascular endothelial cells were blocked by pretreatment with ERK inhibitor (PD98059). In conclusion, the activation of ERK is required for Cronobacter sakazakii invading into brain microvascular endothelial cells.
