摘  要:[目的]探讨异叶天南星氯仿萃取物对肝癌HepG-2细胞的凋亡作用.[方法]采用MTT法测定不同质量浓度的异叶天南星氯仿萃取物对肝癌HepG-2细胞生存抑制率的影响,并利用HE染色法观察各组肝癌HepG-2细胞形态学的变化.[结果]MTT检测结果显示,异叶天南星氯仿萃取物可明显抑制肝癌HepG-2细胞的增殖(P<0.05,P<0.01),且抑制作用随给药时间的延长及药物质量浓度的增加而明显增强;当异叶天南星氯仿萃取物质量浓度为100mg/L,作用时间为48h时,对肝癌HepG-2细胞的抑制率为54.33%,接近IC50.HE染色结果显示,经异叶天南星氯仿萃取物处理的HepG-2细胞表现出细胞间距增大,细胞核固缩、深染,细胞浓缩、碎裂、形成凋亡小体等一系列凋亡形态,且凋亡程度随药物质量浓度的增加及给药时间的延长而增强.[结论]异叶天南星氯仿萃取物可诱导肝癌HepG-2细胞凋亡.OBJECTIVE To explore the apoptotic effects of Arisaema heterophyllum Blume chloroform extract(AHBCE) on hepatoma cell line HepG-2. METHODS The viability inhibitory rate of different concentration of AHBCE on HepG-2 cells was detected by MTT assay, and the effect of AHBCE on morphologic changes of HepG-2 cells was detected by HE staining. RESULTS MTT assay results showed that the AHBCE significantly inhibited the proliferation of HepG-2 cells (P 〈 0.05, P 〈 0.01), inhibitory effect was enhanced with the extension of drug administration time and the increasing drug concentration. As 100 mg/L of AHBCE concentration and 48 h of administration time, the inhibition rate of HepG-2 cells was 54.33 % which was closed to the half maximal inhibitory concentration (IC〈0). HE staining showed that HepG-2 cells treated with AHBCE had larger cell spacing, karyopyknosis, darkened staining, cell fragmentation and the apoptosis bodies, and the apoptotic degree was more serious with the increasing of administration time and drug concentration. CONCLUSION The AHBCE could induce the apoptosis of hepatoma cell line HepG-2 cells.
