摘  要:目的观察葡萄籽原花青素(GSPE)对放射性脑损伤大鼠学习能力和细胞外信号调节激酶1/2(ERK1/2)活性的影响。方法 120只雄性Wistar大鼠随机分为对照组,模型组,U0126组,低、高剂量GSPE组。用直线加速器进行脑部照射22Gy制作放射性脑损伤模型。HE染色观察海马区神经细胞形态变化;免疫组织化学法及免疫印迹法检测磷酸化的ERK1/2表达;穿梭箱评测大鼠学习能力。结果与模型组比较,U0126组海马区神经细胞结构损伤加重,磷酸化ERK1/2表达水平降低(P<0.01),穿梭箱评测显示GSPE组动物主动回避反应率降低,被动回避潜伏期延长(P<0.01);GSPE组海马区神经细胞结构损伤减轻,磷酸化ERK1/2表达水平增高(P<0.01),穿梭箱评测显示GSPE组动物主动回避反应率明显升高,被动回避潜伏期缩短(P<0.01);上述变化在高剂量GSPE组最为显著(P<0.01)。结论 GSPE可以改善放射性脑损伤大鼠学习能力障碍,增强ERK1/2活性。Objective To observe the effects of grape seed proanthocyanidin extract(GSPE)on learning ability and extracellular signal regulated kinase(ERK1/2)activity after radiation-induced injury to the brain in rats.Methods Totally 120 male Wistar rats were divided into control group,model group,U0126 group,and high-and low-dose GSPE groups.The radiation-injured brain models were established by using linear accelerator irradiation in22 Gy.The morphological changes of the neurons in the hippocampus were observed with HE staining.The expression of phosphorylated ERK1/2 was detected by immunohistochemistry and Western blotting.The learning ability was assessed with shuttle box.Results Compared with the model group,U0126 group had increased degree of nerve cell morphological injury and decreased phosphorylated ERK1/2 expression level(P〈0.01).Shuttle box testing showed that the active avoidance reaction rate was decreased and passive avoidance latency was prolonged in GSPE groups(P〈0.01);the GSPE groups had decreased degree of nerve cell morphological injury and increased phosphorylated ERK1/2 expression level(P〈0.01).Shuttle box testing showed that the active avoidance reaction rate was increased and passive avoidance latency was shortened in GSPE groups(P〈0.01).The above-mentioned changes were more significant in high-dose GSPE group(P〈0.01).Conclusion GSPE can reduce learning disability of radiation-injured brain and promote the ERK1/2 activity.
