摘 要:目的观察化痰逐瘀汤对阿尔茨海默病(Alzheimer disease,AD)模型大鼠海马组织丙二醛(MDA)、超氧化物歧化酶(SOD)、β-淀粉样蛋白基因(β-APPm RNA)的影响。方法 2012年9月—2013年9月取10月龄以上Wistar型雄性大鼠,通过海马内注射Aβ25-35制备AD模型,造模成功44只随机分为中药组12只、西药组11只、模型组11只,另选假手术大鼠14只作为空白组。中药组给予化痰逐瘀汤灌胃,西药组给予盐酸多奈哌齐灌胃,空白组及模型组给予等量生理盐水灌胃,各组均干预6周。ELISA法测定海马内MDA、SOD活性变化情况,实时荧光定量PCR技术检测β-APPm RNA转录表达。计量资料采用方差分析,两两比较采用LSD-t检验,P<0.05为差异有统计学意义。结果空白组、模型组、西药组、中药组MDA、SOD水平分别为(5.99±0.52)、(9.27±0.80)、(6.59±0.03)、(6.61±0.03)μmol/g、(205.61±5.89)、(172.95±10.16)、(192.35±9.40)、(194.46±8.51)U/g,四组比较差异均有统计学意义(均P<0.05)。模型组MDA水平高于空白组,SOD水平低于空白组,中药组及西药组MDA水平均低于模型组,SOD水平均高于模型组,差异均有统计学意义(均P<0.05)。空白组、模型组、西药组、中药组β-APPm RNA相对表达水平分别为(1.34±0.30)、(2.53±0.24)、(1.71±0.51)、(1.62±0.40),四组比较差异有统计学意义(P<0.05)。模型组β-APPm RNA相对表达水平高于空白组,中药组及西药组β-APPm RNA相对表达水平均低于模型组,差异均有统计学意义(均P<0.05)。结论化痰逐瘀汤可降低自由基水平,抑制β-APPm RNA的转录,为其治疗老年性痴呆的机理之一。Objective To observe the Huatanzhuyu decoction of Alzheimer' s disease (AD) model rats hippoeampal tissue malondialdehwie (MDA), superoxide dismutase (SOD), β-amyloid protein gene ([3-APPmRNA) impact. Method September 2012 to September 2013 to take 10 months or more type male Wistar rats by injection Aβ25-35 hippocampus preparation AD model, suecessful model 44 were randomly divided into TCM group (n=12), western medicine group (n=11) , model group(n=11), ahernatively sham-operated rats as the control group(n=14). The rats in TCM group were gavageed by Huatanzhuyu decoction, rats in western medicine group were given donepezil hydrochloride orally, rats in control group and model group were given the same amount of normal saline, in each group intervention for 6 weeks. The changes of MDA and SOD aetivity in hippoeampus were measured by ELISA method, real time fluorescent quantitative PCR technique to detect the expression of β-APPmRNA transcription, Measnrement data was processed by ANOVA, P〈0.05 was considered statistically significant. Results The MDA, SOD levels of control group, model group, western medicine group and TCM group were (5.99 ±0.52), (9.27± 0.80), (6.59 ± 0.03), (6.61 ±0.03) μmol/g, (205.61± 5.89), (172.95 ±10.16), (192.35±9.40), (194.46 ± 8.51)U/g, the diffrzeences of the four groups were statistically significant(all P〈0.05).
