摘 要:本文探讨了光照强度和温度2个生态因子对雨生红球藻(Haematococcus plivialis)生长、虾青素及内源脱落酸积累的影响。研究表明,光照强度、温度及二者交互作用对雨生红球藻的生长、虾青素及内源脱落酸的积累均有显著影响(P<0.05)。温度23℃时,细胞密度随光照强度的升高呈先上升后下降趋势,在200μmol/(m^2·s)时达到最大值4.9×105个/mL。适当的高光照强度和高温均能促进雨生红球藻中虾青素和内源脱落酸的积累,过高则起抑制作用。培养期间雨生红球藻中虾青素和内源脱落酸的最大峰值分别为6.266mg/L和322.641ng/g(干重),且最大峰值均出现于27℃、200μmol/(m^2·s)处理组。脱落酸含量的最大值出现在虾青素快速积累前,且在雨生红球藻的生活周期中,孢囊形成期的内源脱落酸含量最高。本研究所得到的规律对规模化培养雨生红球藻具有一定的指导意义。The effects of light intensity and temperature on Haematococcus plivialis growth and accu- mulation astaxanthin and endogenous ABA were determined in this study aimed to a identify the endoge- nous ABA of H. plivialis responding to environmental stress, determine the relationship between the generation of endogenous ABA and astaxanthin accumulation, thus providing a theoretical basis to the large-scale production of H. plivialis. We picked out the alga at exponential growth phase, inoculated it into 2 500 mL of sterile MCM medium(initial density 1X 10^5 cells/mL), cultured the alga under differ- ent continuous light intensities and temperatures for 16days with aeration. According to the results of preliminary experiments and the present comprehensive study, we designed a two-factor cross experi- ment, each experimental factor is divided into three levels, and each processing group is set up three repetitions. The contents of chlorophyll, astaxanthin and carotenoid were determined by spectropho- tometry, and the content of endogenous ABA was measured by enzyme-linked immunosorbent assay. The results showed that the light intensity, temperature and their interaction had significant effects on algal growth and accumulation of astaxanthin and endogenous ABA (P〈0. 05). At 23 ℃, the cell den- sity showed a downward trend after the first rise with the intensity of illumination increased. When light intensity is 200μmol/(m2 · s), the cell density is the largest, and the maximum value is 4. 9X10^5 cells/ mL. In addition, making an appropriate increase in light intensity or temperature can promote the accu- mulation of astaxanthin and endogenous ABA, however, the excessive intensity can inhibit them. Ex- periments explained that the maximum contents of astaxanthin and endogenous ABA were observed at 27 ℃ and 200μmol/(m2 · s), which were 6. 266 mg/L and 322.64 ng/g(dw), respectively.
