尿路上皮癌中Caveolin-1的表达与ERK1/2的相关性研究

摘　　要：目的观察Caveolin-1在正常尿路上皮组织及尿路上皮癌中的表达情况,初步探讨其与ERK信号通路的关系。方法(1)免疫组织化学及RT-PCR检测正常尿路上皮组织及尿路上皮癌中Caveolin-1蛋白及m RNA的表达情况。(2)Western blot方法检测Caveolin-1阳性组及阴性组中ERK1/2及P-ERK1/2的表达。结果尿路上皮癌中Caveolin-1蛋白及m RNA的表达显著高于正常尿路上皮组织,两组相比差异具有显著统计学意义(P<0.05);Western blot结果显示,Caveolin-1阳性组P-ERK1/2的相对表达量显著高于阴性组,而ERK1/2表达在两组之间差异无统计学意义。结论 Caveolin-1的表达上调可能通过激活ERK信号通路而发挥作用,有望为尿路上皮癌的预防和治疗提供新的靶点。Objective To investigate the expression of Caveolin-1 in Urothelium carcinoma and normal urothelium,and discuss the relationship between Caveolin-1 and ERK signal pathway. Methods（1） SP immunohistochemical and RT-PCR were used to detect the expression of Caveolin-1 in urothelium carcinoma and normal urothelium.（2） Western blot analysis were used to detect the expression of ERK1/2 and P-ERK1/2 in Caveolin-1 positive cases and negative cases. Results The expression of Caveolin-1 in urothelium carcinoma was significant higher than that in normal urothelium.（P 0.05）. Western blot revealed that The expression of P-ERK1/2 was significant higher in Caveolin-1 positive cases than that in negative cases. The relative expression levels of AKT in Caveolin-1 positive cases and negative cases were not statistically significant. Conclusion Expression of cavelion-1through activation of ERK1 /2 signaling pathway may play an important role in the process of urothelium carcinoma; Cavelion-1 may be useful targets for prevent and treatment of urothelium carcinoma.
