表柔比星对膀胱癌细胞中miR-21表达的影响

摘　　要：目的表柔比星是膀胱灌注辅助治疗膀胱癌的常用药物,该研究主要通过研究表柔比星对膀胱癌细胞中miR-21表达的影响,从而探讨表柔比星治疗膀胱癌的作用机制。方法该研究通过向膀胱肿瘤细胞中加入不同浓度表柔比星培养细胞后,检测细胞中miR-21表达量的变化情况。结果随着表柔比星浓度的增加,膀胱癌细胞5637中的miR-21的表达水平下降。观察组不同浓度与对照组比较差异有统计学意义(P<0.05),随着表柔比星浓度的增加,人膀胱癌5637细胞中的miRNA-21的表达水平下降,即呈剂量依赖趋势。结论表柔比星能够抑制膀胱癌细胞中miR-21的表达,并呈浓度依赖趋势。Objective Epirubicin is a commonly used drug for assisted bladder perfusion treatment of bladder cancer. This study is to explore the mechanism of epirubicin in the treatment of bladder cancer by studying the effect of epirubicin on the expression of miR-21 in bladder cancer cells. Methods The expressive levels of miR-21 were detected by adding different concentrations of epirubicin in cultured cells to bladder cancer cells. Results The expression level of miR-21 in bladder cancer cell 5637 was decreased with the increase of epirubicin. The difference was statistically significant between the observed groups with different concentrations and the control group（ P〈0. 05）. With epirubicin concentration increased,the expression level of miRNA-21 in human bladder cancer 5637 cells decreased,i. e. in a dose dependent trend. Conclusions Epirubicin can inhibit the expression of miR-21 in bladder cancer cells,which shows a dose-dependent trend.
