DYS452基因座在延边地区朝鲜族和汉族群体中的遗传多态性研究

摘  要:[目的]研究STR基因座DYS452在延边地区朝鲜族和汉族人群中的遗传多态性.[方法]设计DYS452荧光标记引物,利用PCR扩增与ABI310基因分析仪对无关联的各200份朝鲜族和汉族个体的DYS452基因座进行检测,用GeneMapperID v3.2软件分析电泳结果并分析获得群体遗传多态性数据.[结果]朝鲜族和汉族人群在DYS452基因座上共检测出7个等位基因,朝鲜族人群中检测出5个等位基因,分别为29~33号等位基因;汉族人群中检测出7个等位基因,分别为27~33号等位基因.朝鲜族和汉族人群的GD值分别是0.660 7和0.696 4.将DYS452基因座在朝鲜族和汉族中的遗传分布分别与不同地区、民族及种族进行比较,获得不同群体间的P值和遗传距离.[结论]DYS452基因座在延边地区朝鲜族和汉族群体中具有较高的遗传多态性,可为该地区的法医学个人识别、亲权鉴定及人类遗传学调查提供依据.OBJECTIVE To study the genetic polymorphism of the short tandem repeat (STR) locus DYS452 in Chinese Korean and Han nationality in Yanbian area. METHODS Fluoresce labeling primers of DYS452 loci were designed, DYS452 loci were detected in 200 unrelated Chinese Korean and 200 unrelated Han population by PCR amplification and ABI310 genetic analyzer, GeneMapperID v3.2 software was used to analyze the electrophoresis results, and genetic polymorphism data were obtained after analysis. RESULTS 7 alleles were detected from Chinese Korean and Han population in DYS452 loci. 5 alleles were detected from Chinese Korean population, they are No. 29 - 33 allele; 7 alleles were detected from Han population, they are No. 27 - 33 allele. The Gene Diversity values of DYS452 loci in two population were 0. 660 7 and 0. 696 4. P-value and genetic distance were obtained by comparing the genetic distribution of DYS452 loci in Chinese Korean and Han population with other nationality in the different areas. CONCLUSION DYS452 loci have high genetic polymorphism in Chinese Korean and Han population in Yanbian area. The data of DYS452 loci can provide the basis for forensic individual identification, paternity identification and human genetics survey.
