不同低温条件下微管稳定对大鼠供心冷藏损伤的影响

摘  要:为研究不同低温条件下微管改变对大鼠供心保存冷缺血损伤的影响,将36只SD大鼠随机分成对照组(HTK液保存)和实验组(HTK液中含紫杉酚,浓度为10-6 M)。每组的18只SD大鼠再分成4℃、15℃和20℃条件下的3个不同低温保存组,每组6只,低温保存8 h,每间隔2 h灌注5 min。利用离体心脏非循环式Langendorff灌注模式装臵,观察经上述不同保存温度下保存8h后,复温再灌时的冠脉漏出液中心肌酶的含量,并切取心肌组织做光镜和电镜观察。结果显示随着保存温度升高,实验组和对照组中心肌细胞损伤加重,漏出酶增多;与对照组比较,4℃、15℃实验组复温再灌时的冠脉漏出液中乳酸脱氢酶含量明显减少,心肌细胞损伤较轻。说明4-15℃深度低温条件下微管稳定剂能够减轻心肌的冷缺血损伤。To investigate the effect of microtubule changes on the cold storage damage during rat donor's heart preservation in different hypothermy. 36 SD rats were divided randomly into 2 group, the control group, in which heart were arrested and stored in HTK solution, and the experiment group in which HTK solution contained taxol(10-6 M).Then every group was subdived into three subset, namely the isolated rat hearts were stored in 4℃ or 15℃or 20℃ for 8 hours, every 2 hours interval to perfuse for 5 min. With Langendorff model perfusion apparatus, isolated rat hearts were stored in different temperature for 8 hour, Then the myocardial enzyme leakage in coronary effluent were detected, the heart sample were obtained and observed under light and electron microscopes after reperfuse. The result shows that the higher preserve temperature, the more damage and the more leakage of enzyme have taken place. Compared with control group, the leakage of lactate dehydrogenase(LDH) was significantly reduced and cell damage was lessened in 4℃ or 15℃ experiment groups. There for we believe that microtubulin stabilizing agent can lessen the cold storage damage of cadiocyte in the 4-15 ℃ deep hypothermia preservation.
