摘  要:目的探讨体外1,25(OH)2D3对糖尿病视网膜病变(Diabetic Retinopathy,DR)单个核(Peripheral Blood Mononuclear Cells,PBMCs)细胞增殖和分泌细胞因子的影响。方法选择维吾尔族增殖期DR患者20例(PDR组),非增殖期DR患者24例(NPDR组)、正常对照22例(NC组),分离受试者外周血PBMCs细胞,体外1,25(OH)2D3刺激后用CCK8法检测细胞增殖;分离PBMCs后加入抗CD3/CD28抗体活化,加或不加1,25(OH)2D3培养,收集细胞培养上清并使用Cytometric Bead Array技术检测上清中细胞因子的水平。结果 1,25(OH)2D3能显著抑制DR患者和正常对照的PBMCs细胞增殖,差异有统计学意义(P<0.05),并且1,25(OH)2D3能抑制DR患者和正常对照的PBMCs分泌IL-17A、IFN-γ和IL-6因子,差异有统计学意义(P<0.05)。结论 1,25(OH)2D3作为一种免疫调节剂,对DR患者免疫细胞的增殖和相关细胞因子的分泌具有抑制作用。Objective To investigate the effect of 1,25(OH)_2D_3 on the proliferation and secretion of cytokines in the Peripheral blood mononuclear cells(PBMCs)from diabetic retinopathy(DR).Methods The subjects were collected from 20 proliferative DR,24 non proliferative DR and 22 normal controls in our hospitals.Peripheral blood mononuclear cells(PBMCs)were cultured with or without 1,25(OH)_2D_3 to detect the change of proliferation.The Cell Counting Kit was used to determine the rate of cell proliferation.PBMCs were cultured with or without 1,125(OH)_2D_3 in the presence of anti-CD3 and anti-CD28 to detect the change of secretion of cytokines.The Cytometric Bead Array system was used to analyse cytokine levels in the culture supernatant.Results 1,25(OH)_2D_3 significantly inhibited the proliferation of PBMCs and the secretion of IL-17 A,IFN-γand IL-6cytokines(P 0.05).Conclusion 1,25(OH)_2D_3 may be play as an immune modulating agent to suppress cell proliferation and related cell cytokines among DR patients.
