摘 要:为了探讨猪精子微胶囊制作参数,优化猪精子微胶囊制作工艺,提高微胶囊及微胶囊化保存的猪精液质量。将猪精液进行稀释变成1×10^8个/m L,利用海藻酸钠和钡离子反应制成猪精子微胶囊,检测海藻酸钠和钡离子不同反应时间对胶囊的参数及猪精子质量的影响,以确定最优的微胶囊反应时间的参数。结果表明:5、20、60 min反应时间组之间在胶囊强度分别为23.5、35.5、66.0 g/个,壁厚分别为557.5、1 009.0、1 156.5μm,重量分别为80.0、112.0、135.0 mg以及缓释率(24 h时分别为9.40×104、6.00×10^4、4.50×10^4)sperm/个胶囊,以上指标存在显著差异(P〈0.05)。其中5 min反应组的强度最差;而60 min反应组缓释率最低,不利于精子的释放。此外,20 min和5 min反应时间组在微胶囊化保存后猪精子的活力显著高于60 min反应组,而在精子畸形率上都明显低于60 min反应时间组(P〈0.05)。综上可见,钡离子-海藻酸钠微胶囊反应时间在20 min时达到最佳,微胶囊保存猪精子质量最好。The main purpose of th is study was to investigate the parameters of microcapsules for Boar semen and optimize production process of boar semen microcapsule, to improve the quality of microcapsule for boar semen. The boar semen was diluted to 1 ×10^8 cells/ml by diluent, we used sodium alginate and barium ion reaction to make microcapsule, and to detect the effect of different reaction time on parameters of capsule and quality of boar sperm. Results showed that there were significant differences in the strength of the capsules 23.5 vs. 35.5 vs. 66.0 g/one capsule, wall thickness 557.5 vs. 1009.0 vs. 1156.5 p,m, respectively, weight 80.0 vs. 11.2 vs. 1.35 rag,respectively), and Slow release rate 9.40×10^4 vs. 6.00×10^4 vs. 4.50±0.30×10^4 sperm/one capsule for 24 h) in three groups(P〈0.05). Among them, the strength of 5min group was the worst, while the release rate of 60min group was the lowest in all groups, which was not conducive to the release of sperm. In addition, the sperm viability of the 20min and 5rain group were significantly higher than that of the 60 rain group, and the rate of sperm abnormality was significantly lower than t the 60 min group (P〈0.05). This study's results confirmed sodium alginate and barium ion reaction time for 20min was suitable for the production of boar sperm microeapsules.
