摘 要:目的 观察Slit同源蛋白2(Slit2)及其受体跨膜受体蛋白1(Robo1)在脑胶质瘤中的表达及其在胶质瘤血管新生中的作用.方法 应用免疫组织化学技术检测10个样本的正常脑组织及55个样本的低、高级别胶质瘤组织中的表达,同时对肿瘤内微血管密度(MVD)进行检测,比较Slit2/Robo1在不同组别中的表达强度,并分析其与MVD的关系.结果 与正常脑组织比较,Slit2在胶质瘤中的表达强度下降,且肿瘤恶性程度越高,Slit2的表达强度越低,甚至完全不表达[正常脑组织组、低级别组胶质瘤和高级别胶质瘤组表达强度为(+++)和(++++)的百分率分别为100.00%(10/10)、69.23%(18/26)、10.34%(3/29)].Robo1的表达强度随胶质瘤的恶性程度增高而增高[正常脑组织组、低级别组胶质瘤和高级别胶质瘤组表达强度为(+++)和(++++)的百分率分别为0.00%(0/10)、84.62%(22/26)、93.10%(27/29)].在正常脑组织、低级别胶质瘤、高级别胶质瘤3组之间的Slit2、Robo1的表达强度及MVD计数差异均有统计学意义(P<0.05).在所检测的所有3组样本中,Slit2和Robo1的表达呈负相关(r=-0.743,P<0.05),Slit2的表达与MVD呈负相关(r=-0.751,P<0.05),而Robo1的表达与MVD呈正相关(r=0.859,P<0.05).结论 Slit2可通过其信号通路抑制胶质瘤新生血管的形成,进而抑制胶质瘤发生、迁移、浸润。Objective To investigate the expression of slit homolog 2 protein (Slit2) and its receptor roundabout 1 (Robo1) in the glioma,and their role in glioma angiogenesis.Methods Immunohistochemical technique was used to detect the expression of Slit2/Robo1 from 10 samples of normal brain tissues and 55 samples of low and high grade glioma tissues,and to compare the expression levels in different groups.Meanwhile,the intratumoral microvessel density (MVD) of all the 55 samples was also tested to analyze its relationship with Slit2/Robo1.Results As compared with normal brain tissues,the expression of Slit2 was lowered in the glioma tissues,and the higher of the tumor malignant degree,the lower of the expression of Slit2,even no expression [normal brain tissue and low-grade group of glioma and high-grade glioma group expression intensity percentage of (+++) and (++++) respectively:100.00% (10/10),69.23% (18/26),10.34% (3/29)].Robo1 expression increased with malignant degree of gliomas [normal brain tissue and low-grade group of glioma and high-grade glioma group expression intensity percentage of +++and +++ respectively:0.00% (0/10),84.62% (22/26),93.10% (27/29)].Among the normal brain tissues,low grade gliomas,and high grade gliomas,the expression of Slit2 and Robo1 and MVD counting showed statistically significant difference (P 〈 0.05).In the three groups of samples,the expression of Slit2 and Robo1 was negatively correlated (r =-0.743,P 〈0.05P);the expression of Slit2 was negatively correlated to the MVD (r =-0.751,P 〈 0.05);while the expression of Robo1 was positively correlated with MVD (r =0.859,P 〈 0.05).Conclusion Slit2 can restrain the formation of new blood vessels through the signal pathways,thereby inhibiting glioma formation,migration,and infiltration.
