摘 要:目的:研究非小细胞肺癌(non-smallcelllungcancer,NSCLC)患者血清组蛋白去乙酰化酶1(histonedeacetylases1,HDAC1)和DNA甲基化转移酶1(DNAmethyltransferase1,DNMTl)含量与肿瘤临床病理分期、恶性分子表达的相关性。方法:选择2012年4月~2015年10月在我院确诊为NSCLC的89例患者作为恶性组,并收集血清样本、肺癌组织,64例肺部良性病变患者作为良性组并收集血清标本,测定血清中HDAC1、DNMT1含量以及肺癌组织中p53、p21、PTEN、C-myc、MKi-67的表达量。结果:恶性组血清中DNMT1和HDAC1的含量显著高于对照组(P〈0.05),且TNM分期越高、血清中DNMT1和HDAC1的含量越高,而不同组织类型NSCLC患者血清中DNMT1和HDAC1的含量无显著性差异(P〉0.05);恶性组TNM分期越高,肿瘤组织中p21、p53、PTEN的表达量越低,且与血清DNMT1、HDAC1含量呈负相关,C-myc、MKi-67的表达量越高且与血清DNMT1、HDAC1含量呈正相关。结论:NSCLC血清中DNMT1和HDAC1的含量显著升高并且能够降低抑癌基因表达、促进原癌基因表达并参与肿瘤的发生发展。Objective: To study the correlation of serum HDAC1 and DNMT1 levels with the clinical pathological stag- ing and malignant molecule expression in patients with NSCLC. Methods: A total of 89 patients diagnosed with NSCLC in our hospital from April 2012 to October 2015 were selected as malignant group, serum samples and lung tissues were collected from them, 64 patients with benign lung lesions were selected as benign group, serum specimens were collected from them, and then the HDAC1 and DNMT1 levels in serum as well as p53, p21, PTEN, c-myc and MKi-67 expression levels in lung tis- sue were determined. Results.. DNMT1 and HDAC1 levels in serum of malignant group were significantly higher than those of control group, the higher the TNM staging, the higher the serum DNMT1 and HDAC1 levels, and serum DNMT1 and HDAC1 levels were not significantly different among patients with different tissue types of NSCLC~ the higher the TNM stag-ing of malignant group, the lower the p21, p53 and PTEN expression levels in tumor tissue and they were negatively correlated with DNMT1 and HDAC1 levels in serum, and the higher the c-myc and MKi-67 expression levels and they were positively correlated with DNMT1 and HDAC1 levels in serum. Conclusions: Serum DNMT1 and HDAC1 levels in patients with NSCLC significantly increase and can lower tumor--suppressor gene expression, promote proto-oncogene expression and participate in the occurrence and development of tumor.
