摘 要:目的:探讨Bmi-1基因在口腔鳞癌组织中的表达及意义。方法:收集口腔鳞癌(OSCC)患者手术切除新鲜标本30例作为OSCC组,癌旁0.5 cm外正常组织及正常体检者的口腔黏膜组织共30例作为正常对照组,免疫组织化学染色SP法检测两组组织中Bmi-1蛋白的表达,实时定量基因扩增荧光检测系统(QPCR)检测Bmi-1 mRNA的表达,比较不同临床理特征OSCC患者Bmi-1的表达差异。结果:OSCC组织中,Bmi-1蛋白和mRNA的表达明显高于正常对照组;在OSCC组织中,中低分化的OSCC组织的Bmi-1蛋白和mRNA表达高于高分化OSCC组织,差异具有统计学意义(P<0.05),Bmi-1表达与患者年龄、性别无关(P>0.05)。结论:Bmi-1基因检测可作为OSCC的辅助诊断指标之一。Objective: To explore the Bmi-1 gene expression and clinical significance in oral squa- mous carcinoma(OSCC) tissues. Methods: Fresh tissue samples of 30 patients with oral squamous cell carcinomas were collected as OSCC group, while 30 cases of 0.5 cm normal tissue adjacent to car- cinoma and normal physical examination were taken as the normal control group. The expression of Bmi-1 protein in the two groups was detected by immunohistochemistry SP method, and the expression of mRNA Bmi-1 was detected by real-time quantitative PCR. Results: The expression of Bmi-1 protein and mRNA in OSCC tissue were significantly higher than those in normal control group. In OSCC tis- sues, the expression of Bmi-1 protein and mRNA in the low and middle differentiated OSCC tissues were higher than those in the highly differentiated OSCC tissues, and the difference was statistically significant ( P 〈 0.05). BMI-1 expression was not related to patients age and sex. Conclusion : Broi-1 gene detection can be used as one of the auxiliary diagnostic indicators of oral squamous cell carcino mas.
