摘  要:目的探讨mi R-1297在他莫昔芬(TAM)介导的子宫内膜癌中的表达及临床意义。方法收集2012年3月1日至2015年3月1日期间首都医科大学附属北京天坛医院乳癌术后服用TAM超过5年的可疑内膜病变患者10例,对照组选用因子宫脱垂而行子宫切除的患者,没有激素和TAM服药史,HE染色确定病理类型。离体培养子宫内膜原代细胞,用TAM处理后采用细胞克隆方法检测细胞的生长情况,及Real time-PCR方法检测组织及细胞中mi R-1297的表达。结果对照组、服用TAM的非典型增生组、子宫内膜癌组中mi R-1297表达呈递增趋势。此外,离体培养的子宫内膜细胞,用TAM处理后发现细胞增殖,检测mi R-1297表达上调。结论在TAM介导的子宫内膜癌变中mi R-1297促进细胞生长,具有致癌作用。这能让我们更好地理解TAM介导子宫内膜癌的机制。对TAM介导的子宫内膜癌而言,mi R-1297可能是一个潜在的早期临床预测的生物标志物。Objective To investigate the expression and clinical significance of mi R-1297 in tamoxifen(TAM) mediated endometrial carcinoma. Methods 30 patients with breast cancer undergoing surgical therapy and taking TAM over five years were enrolled during 2012-2015 in Beijing Tiantan Hospital. The patients underwent hysterectomy due to uterine prolapse without hormones or TAM taken history were enrolled as control group. HE staining was used to identify the type of pathology. Primitive cells of endometrium were cultured in vitro. After TAM exposed, cell growth was detected by cell cloning, and the expression of mi R-1297 in the tissues and cells was detected by Realtime-PCR method. Results Mi R-1297 expression showed an increasing trend in the control group, taking TAM atypical hyperplasia, and endometrial carcinoma. Moreover, after exposed with TAM, cell proliferation and mi R-1297 were upregulated in normal endometrial cells. Conclusions Mi R-1297 promoted cell growth and had a carcinogenic effect in TAM-mediated endometrial. Mi R-1297 could be a potentially clinical predictive biomarker to TAM mediated endometrial carcinoma.
