摘  要:TAPBP是抗原呈递相关转运体(TAP)相关蛋白,在先天性和适应性免疫反应中均发挥着重要作用,与多种疾病的抗性或易感性相关。对25个不同品种鸡TAPBP基因序列进行目标捕获测序,利用MEGA软件对不同品种鸡序列进行比对分析,筛选不同品种鸡TAPBP基因上的SNPs及Indel位点,并根据基因序列进行系统进化树的构建。结果表明:TAPBP基因存在较多的SNPs和Indels位点,有着丰富的遗传多样性。不同地方鸡品种相对外来鸡品种呈现出高度多态性,可能与抗病性存在密切联系。青脚麻鸡、太湖鸡、如皋草鸡等抗性较好的鸡品种在系统进化树中拥有较高的亲缘系数。此外,固始鸡、如皋草鸡、安卡鸡等品种在第8外显子上存在特异性的插入缺失,且引起氨基酸变化,可作为鸡抗性研究的候选分子标记。TAPBP as the transporter associated with antigen processing (TAP) binding protein, plays an important role in adaptive and congenital immune response, which caused disease resistance or susceptibility. This study conducted the target capture sequencing for 25 chicken breeds. Different breeds sequences were alimented by MEGA software, then screened out SNPs and Indels of TAPBP gene in different chicken breeds and constructed phylogenetic tree. The result showed that TAPBP gene had a lot of SNPs and Indels, and had rich sequence polymorpbism. Compared with the foreign chicken, Chinese local chicken breeds showed high polymorphism, which might be closely linked with disease resistance. Moreover, some breeds with good disease resistance, such as Rucao chicken, Gushi chicken, Anka chicken, had specific indels in the 8th exon, which caused the changes of amino acids and may be taken as a candidate genetic marker with the disease resistance.
