摘  要:目的研究人脑胶质瘤组织中微小RNA-374(microRNA-374,miR-374)的表达及其与临床病理特征之间的关系,探讨其在胶质瘤患者中的临床意义。方法采用RT-PCR法检测85例胶质瘤和30例正常脑组织中的miR-374表达;Kaplan-Meier法分析胶质瘤患者的总生存期;观察慢病毒过表达miR-374载体转染U251和U87细胞株后对肿瘤细胞增殖的影响。结果胶质瘤组织中miR-374的表达明显下调;miR-374的表达与WHO分级(P=0.008)、Karnofsky评分(KPS)(P=0.021)和生存时间(P=0.01)显著相关;Kaplan-Meier分析结果表明,miR-374低表达与高表达患者总生存期(P<0.01)的差异有统计学意义,miR-374低表达的患者预后较差。多因素分析显示胶质瘤中miR-374的差异表达是预测胶质瘤患者预后的独立危险因素(HR:5.75,95%CI:3.56-6.93,P=0.005)。体外实验表明,过表达miR-374可以显著抑制胶质瘤细胞的增殖。结论 miR-374在胶质瘤中是低表达的,与胶质瘤患者预后显著相关;miR-374高表达可抑制胶质瘤细胞的增殖。Objective To investigate the relationship between microRNA-374 (miR-374) expression and clinicopatholog- ic factors in glioma tissues, and to evaluate the clinical significance of miR-374 in glioma patients. Methods The expression lev- els of miR-374 were analyzed in 85 glioma tissues and 30 normal brain tissues by real-time PCR (RT-PCR). Survival analysis by Kaplan-Meier method was performed to assess prognostic significance. And miR-374 over-expression lentiviral vector (Lv-miR- 374) and a control lentiviral empty vector (LEV) were stably transfected into human glioma U251 and U87 cells, to evaluate the ef- fect of niR-374 on cell proliferation. Results Down-regulation of miR-374 was detected in most primary glioma tissues, and which was significantly associated with WHO grade(P=0.008), Karnofsky performance scorc(KPS)(P=0.021 ), and survival time(P=0.01 ).Using Kaplan-Meier analysis, we found that low expression of miR-374 was associated with a worse prognosis. Multivariate analyses showed that miR-374 expression was the independent risk factors for predicting overall survival time of glioma patients. Conclusion MiR-374 is down-regulated in glioma tissues, miR-374 expression is significantly correlated with the prognosis of glio- ma patients, and up-regnlation miR-374 in U251 and U87 cells could inhibit the prolifrration of glioma cells.
