摘 要:目的观察补骨脂-肉豆蔻醇提物对肠易激综合征内脏高敏感大鼠的治疗作用及其可能机制。方法将新生SD大鼠随机分为6组,正常组,模型组,补骨脂-肉豆蔻醇提高、中、低剂量组及匹维溴铵组,每组8只。除正常组外,其余各组采用母子分离结合乙酸灌肠法复制内脏高敏感大鼠模型,于造模后第6周起,治疗组大鼠给予醇提物或匹维溴铵灌胃,连续两周。观察结肠组织病理学变化及内脏高敏感性相关指标,采用酶联免疫吸附法(ELISA)检测P物质(SP)和降钙素基因相关肽(CGRP)含有量,通过免疫荧光法检测促肾上腺皮质激素释放因子(CRF)和CRF1型受体(CRFR1)表达。结果与模型组相比,补骨脂-肉豆蔻醇提物(15、10 g/kg)能够升高大鼠腹部回缩反射阈值,降低大鼠结肠运动和结肠组织中SP、CGRP、CRF、CRFR1的表达。结论补骨脂-肉豆蔻对肠易激综合征内脏高敏感性大鼠有一定的治疗作用,其机制与调节结肠组织中SP、CGRP、CRF及其受体有关。AIM To observe the therapeutic effect of alcohol extracts from scurfpea fruit( Psoralea corylifolia L.) plus nutmeg apple( Myristica fragrans Houtt.) on the visceral hypersensitivity in rats with irritable bowel syndrome and its possible mechanism. METHODS Newborn SD rats were randomly divided into six groups,control,model,low-,middle- and high-dose alcohol extracts from scurfpea fruit plus nutmeg apple,and Pinaverium Bromide Tablets groups,eight rats in each group. Except for the control group,rats in all other groups were separated from their mothers and acetic acid-induced visceral hypersensitivity starting from the sixth week. These rats in the treatment groups were administered with alcohol extracts or Pinaverium Bromide Tablets by gastrogavage for two consecutive weeks. The pathological changes of colonic tissue and visceral hypersensitivity-related indicators were observed. The contents of P substance( SP) and calcitonin gene-related peptide( CGRP) were determined by enzyme-linked immuno sorbent assay( ELISA). The expressions of corticotropin-releasing factor( CRF) and CRF receptor-1( CRFR1) were determined by immunofluorescence technique. RESULTS Compared with the model group,15 g / kg and 10 g / kg alcohol extracts from scurfpea fruit plus nutmeg apple could not only increase the pressure threshold of abdomenal withdrawal reflex,but also decrease the colon movement and the expressions of SP,CGRP,CRF,CRFR1 in colonic tissue in rats. CONCLUSION Alcohol extracts from scurfpea fruit plus nutmeg apple have a certain curative effect on the visceral hypersensitivity in rats with irritable bowel syndrome.Their mechanism of action may be related to the regulation of SP,CGRP,CRF and its receptor level.
