摘  要:目的探讨金欣口服液治疗呼吸道合胞病毒(RSV)肺炎的可能作用机制。方法将48只BALB/c小鼠随机分为正常组、RSV肺炎模型组、金欣口服液组,每组16只。RSV肺炎模型组和金欣口服液组小鼠用RSV悬液滴鼻感染小鼠建立RSV肺炎模型。金欣口服液组予金欣口服液27.6 g/(kg·d)灌胃,正常组和RSV肺炎模型组小鼠给予等量的生理盐水灌胃。连续灌胃至第4天每组处死5只小鼠进行病理学检查,其他小鼠于第6天分别收集尿液,运用GC-MS检测尿液中所含有的代谢物及其相对含量的变化。结果RSV肺炎模型组小鼠肺脏可见间质多灶性炎性浸润,肺泡壁轻度充血,明显增厚。金欣口服液组小鼠肺部炎症较RSV肺炎模型组小鼠肺部炎症明显减轻。RSV肺炎小鼠的尿液中共测得38种代谢产物,其中乳酸、甘氨酸、棕榈酸、硬脂酸为差异性代谢产物,RSV肺炎模型组乳酸、甘氨酸、硬脂酸、棕榈酸相对含量明显高于正常组和金欣口服液组(P<0.05)。结论金欣口服液可能通过调节体内能量代谢、氨基酸代谢、脂肪酸代谢治疗RSV肺炎。Objective To explore the possible functional mechanism of Jinxin Oral Liquid( 金欣口服液) on respiratory syncytial virus( RSV) pneumonia. Methods Forty-eight 48 BALB / c mice were randomly divided into a normal group,a RSV pneumonia group,and a Jinxin Oral Liquid group,with 16 mice in each. The RSV pneumonia group and the Jinxin Oral Liquid group accepted RSV suspension nasal drip to infect mice to build RSV pneumonia model. The Jinxin Oral Liquid group was given Jinxin Oral Liquid 27. 6 g /( kg·d) for gavage. The normal group and the RSV pneumonia group were given saline with equal dose for gavage. Five mice in each group were executed for pathological examination on the fourth day. Urine sample of other mice were collected on the sixth day respectively and GC-MS was adopted to examine urine metabolites and the changes of its relative levels. Results There were mesenchymal multifocal inflammatory infiltrations,slight hyperaemia and remarkable thickness in alveolar wall in the lung of RSV pneumonia model mice. The lung inflammation in the Jinxin Oral Liquid group remarkably decreased compared with that in the RSV pneumonia group. There were 38 types of metabolites examined in urine of mice in the RSV pneumonia group,among which lactic acid,glycine,palmitic acid and stearic acid were variant metabolites. In the RSV pneumonia group,the relative levels of lactic acid,glycine,palmitic acid and stearic acid were remarkably higher than that in the normal group and the Jinxin Oral Liquid group( P 〈0. 05). Conclusion Jinxin Oral Liquid could cure RSB pneumonia by means of regulating energy metabolism,amino acid metabolism and fatty acid metabolism.
