胃癌组织中microRNA-224阳性表达与患者预后相关性分析

摘　　要：目的:探讨micro RNA-224于胃癌组织中的阳性表达与患者临床病理特征及预后的相关性。方法:应用原位杂交技术检测micro RNA-224在50例石蜡固定癌旁非肿瘤胃黏膜组织及112例石蜡胃肿瘤组织切片中的表达情况,分析其与肿瘤患者各病理参数及预后的相关性。原位杂交按照原位杂交试剂盒的说明书进行。结果:micro RNA-224在胃癌组织中阳性表达率49.1%(55/112),癌旁非肿瘤胃黏膜组织中阴性表达。其阳性表达与TNM分期、淋巴结转移、远处转移及浸润深度存在相关性(P<0.05),而与性别、年龄、肿瘤大小、肿瘤的部位、分化程度、Lauren分型无关(P>0.05)。micro RNA-224阴性表达比阳性表达者5年生存率高,差异有统计学意义(P<0.001)。经COX多因素分析显示:micro RNA-224的表达是影响胃癌患者预后的独立因素之一(P<0.05)。结论:micro RNA-224在肿瘤组织中阳性表达与TNM分期、淋巴结转移、远处转移及浸润深度密切相关,是影响胃癌患者预后的独立因素之一。Objective：To explore the correlation of the expression of micro RNA-224 in human gastric cancer tissues and clinical pathological characteristics and prognosis of patients.Method：In situ hybridization was used to detect the expression of micro RNA-224 in 112 paraffin-embbeded human gastric cancer tissue sections and 50 cases of adjacent non-tumor gastric mucosa,and its relationship between various pathological parameters and prognosis of the patients was then analyzed.The procedures performed according to manufacturer＇s instructions of sensitivity-enhanced in situ hybridization kits.Result：Micro RNA-224 was negative in human non-tumor gastric mucosa samples,but was positive in 55 of 112 cases（49.1%） of gastric cancer.Positive expression of micro RNA-224 correlated with the depth of invasion,lymph node metastasis,distantmetastasis and TNM stage（P0.05）,but not correlated with patients＇ age,gender,tumor locationdegree of differentiation,tumor size and Lauren classification（P0.05）.The 5-year survival rate of patients with a positive expression of micro RNA-224 was significantly lower than that in patients with negative expression,the difference was statistically significant（P0.001）.Cox regression analysis showed that the expression of micro RNA-224 was one of the independent prognostic factors for patients with gastric cancer（P0.05）.Conclusion：Positive expression of micro RNA-224 correlates with the depth of invasion of gastric cancer patients,TNM stag,lymph node metastasis and distant metastasis.The expression of micro RNA-224 was an independent prognostic factor for patients with gastric cancer.
