摘 要:目的探讨刺槐素在不同时间点给药对小鼠脑缺血再灌注损伤的保护作用,并分析刺槐素的治疗时间窗。方法将36只雄性昆明小鼠随机分为假手术组、缺血再灌注组、刺槐素(治疗)组,治疗组又根据不同给药时间分为0、2、4、8h 4个给药组,每组各6只;采用线栓法制作大脑中动脉闭塞模型,治疗组予缺血0、2、4、8h腹腔注射刺槐素(12.5 mg/kg);再灌注后24h进行神经功能评分,应用2,3,5-氯化三苯基四氮唑染色测定脑梗死体积。结果缺血0、2、4h给药组神经功能评分均显著低于缺血再灌注组(p均<0.05),8h给药组神经功能评分有改善,但无统计学意义(p=0.052);不同时间给药组间神经功能评分无统计学意义(p>0.5)。缺血再灌注组脑梗死体积为(65.18±5.96)mm^3,与缺血再灌注组相比,缺血0h[(22.15±3.02)mm^3,p=0.000],2h[(28.61±4.19)mm^3,p=0.000],4h[(49.8±4.08)mm^3,p=0.000]给药组脑梗死体积均显著降低,8h[(56.09±7.92)mm^3,p=0.075]给药组脑梗死体积有减小,但无统计学意义。结论在缺血早期应用刺槐素治疗可有效改善神经功能评分,缩小脑梗死体积。Objective To investigate the neuroprotective effects of Acacetin in Mice after cerebral ischemia and reperfusion when it was administered at different time points and analyze acacetin therapeutic time window. Methods Intraluminal suture method was used to develop reversible middle cerebral artery occlusion (MACO). 36 male Kunming mice are randomly divided into sham group, ischemia-reperfusion group, ischemi- a-reperfusion therapy group. The therapy group according to different drug administration time is divided into: 0 h, 2 h, 4 h, 8 h 4 groups(n = 6 in each group). The mice in ischemia-reperfusion therapy group were injected intraperitoneally at 0,2, 4, 8 hours after MACO (12. 5 mg/kg ). The neurological function score was evaluated at 24 hours after reperfusion. Cerebral infarct volume was determined by 2,3,5-triphenyl tetrazolium stai- ning. Results The neurological function score in the groups of 0 h, 2 h, 4 h were lower than that in the ischemi- a-reperfusion group(all p〈0. 05), The neurological function score in 8 h group has no statistical significance. There was no significant difference among the groups which administered at different time points. The infarct volume in the groups of Oh ,2 h,4 h was (22. 15 ±3.02)mm3 , (28. 61± 4. 19)mm3 , (49. 8± 4. 08)mm3 , which was significantly smaller than that in the ischemia-reperfusion group (65.18 ±5.96)mm3, (all p〈0. 05). There Was no significant difference between the 8h group and ischemia-reperfusion group. Conclusion Acacetin can improve neurological function score and reduce the infarct volume in cerebral ischemia early stage.
