摘 要:目的探讨补骨脂素对去卵巢痴呆大鼠脑组织海马区的病理形态学影响。方法 2013年2月~2016年1月制备去卵巢痴呆大鼠模型,分为假手术组、模型组和补骨脂素组,每组10只,其中补骨脂素组建立模型后连续灌胃给药29 d,模型组给予常规生理盐水代替。观察比较各组大鼠脑组织海马区的病理形态。结果与假手术组比较,模型组大鼠潜伏期明显延长(P〈0.05);与模型组比较,补骨脂素组均能明显缩短潜伏期(P〈0.05);补骨脂素组大鼠穿越平台次数显著增加,与模型组比较,差异有统计学意义(P〈0.05)。假手术组、模型组、补骨脂素组SOD含量分别为(145.30±18.42)、(110.49±6.46)、(120.98±7.13)U/gprot,MDA含量分别为(2.87±0.44)、(4.46±0.63)、(3.19±0.42)nmol/mgprot。与假手术组比较,模型组SOD含量显著降低,MDA含量显著增加,三组比较差异有统计学意义(P〈0.05)。超微透射电镜显示补骨脂素组锥体细胞形态得到改善,排列均一、整齐,结构清晰。结论补骨脂素能提高去卵巢痴呆大鼠的SOD氧化酶活性,降低MDA含量,从而有利于脑组织海马区的病理形态学的改善,提高记忆能力。Objective To investigate the effects of psoralen on ovariectomized rat dementia in hippocampus of rat brain tissue pathological effect.Methods From February 2013 to January 2016,the preparation of ovariectomized dementia rat model were divided the sham operation group,model group and psoraleae group,with 10 rats in each group,included psoralen group model after intragastric administration 29 d,model group were given routine saline instead.The pathological changes of the hippocampus of rats in each group were observed and compared.Results Compared with the sham group,rats in the model group,the latency were significantly prolonged(P〈0.05);compared with the model group, psoralen group were markedly shorted the incubation period( P〈0. 05); and at the same time the psoralen group rats across the platform number were increased significantly, and compared the model group were significant difference(P〈0.05).The SOD contents in the sham group, model group,psoraleae group were(145.30 ±18.42),(110.49 ±6.46) and(120.98±7.13)U/gprot,the content of MDA were(2.87 ±0.44),(4.46 ±0.63) and(3.19±0.42)nmol/mgprot.Compared with the sham group,SOD content of the model group were significantly decrea sed,MDA content were increased significantly;and the contents of SOD and MDA in the psoralen group compared to the other groups difference were statistical significance(P〈0.05).Ultrastructural TEM showed that the morphology of pyramidal cells of psoralen group improved,uniform,neatly arranged,clear structure.Conclusion Psoralen in dementia rat ovarian application can improve the SOD enzyme activity, reduce MDA content, thus benefit the brain hippocampus pathological improvement,improve memory ability.
