摘 要:目的以MCHC、MCH和Hb A、Hb A2、Hb F等血象为观察指标,探讨乙基亚硝基脲(ethylnitrosourea,ENU)复制β-地贫模型的可行性。方法实验设模型组和正常对照组两组。模型组:先给雄性DBA/2J小鼠腹腔注射乙基亚硝基脲(200 mg/kg体重),经9周诱发遗传基因突变,然后将其与正常雌性C57BL/6J小鼠杂交繁育出具有β-地中海贫血特征的模型杂交F1代小鼠。正常对照组:将正常雄性DBA/2J小鼠与正常雌性C57BL/6J小鼠杂交繁育出正常杂交F1代小鼠。结果与正常杂交F1代小鼠比较,模型杂交F1代的部分小鼠血MCHC、MCH、Hb A显著降低,Hb A2和Hb F显著升高(P<0.05或P<0.01)与临床初步诊断患者为β-地中海贫血的相应血象指标的变化趋势相一致。结论乙基亚硝基脲可诱发小鼠类似β-地中海贫血。Objective To investigate the feasibility of ethylnitrosourea(ENU)in the establishment of mouse model of β-thalassemia using blood markers MCHC,MCH,Hb A,Hb A2 and Hb F as observational indicators. Methods DBA/2J micewere randomly divided into model group and normal control group. In the model group,model hybrid F1 mice were bred by ENU(200 mg/kg,for 9 weeks)-induced male DBA/2J mice and normal female C57BL/6J mice. In the normal group,the normalhybrid F1 mice were bred by normal male DBA/2J mice and normal female C57BL/6J mice. Results Compared with thenormal hybrid F1 mice,some mice from the model hybrid F1 mice showed significantly decreased MCHC,MCH and Hb A,andobviously increased Hb A2 and Hb F(P0.05 or P0.01),which were consistence with the changes in the freshly diagnosed β-thalassemia patients. Conclusion ENU is liable to induce β-thalassemia-like condition in mice.
