散发性乳癌组织BRCA1和BRCA2蛋白表达及意义

摘　　要：目的研究散发性乳癌组织中乳癌易感基因1（BRCA1）与BRCA2蛋白的表达及其临床意义。方法选取本院乳腺外科手术切除后经病理证实的乳癌病人的乳癌组织74例,癌旁组织40例,同时选取乳房增生症组织40例,采用免疫组织化学方法检测3组标本中BRCA1、BRCA2蛋白的表达情况,并分析其临床意义。结果乳癌组织中BRCA1、BRCA2蛋白阳性表达率均显著低于乳房增生组织、癌旁组织,差异有显著性（χ~2=24.63~27.53,P〈0.05）。乳癌组织中BRCA1、BRCA2蛋白的表达与病人的年龄、肿瘤最大径、组织学分级、淋巴结转移、病理类型无关（P〉0.05）。结论 BRCA1、BRCA2蛋白表达下调可能是散发性乳癌发病的一个重要因素,但两者与病人临床病理特征的关系不明显。Objective To study the expressions of BRCA1 and BRCA2 protein in breast cancer tissue and its clinical significance. Methods Breast cancer tissue of 74 cases confirmed by postoperative pathology,paraneoplastic tissue of 40 cases,and tissue of 40 cases of breast hyperplasia were collected for this study.Employing immunohistochemical method,the expressions of BRCA1 and BRCA2 were detected and their clinical significance analyzed. Results The expressions of BRCA1 and BRCA2 in breast cancer tissue were markedly lower than that in paraneoplastic tissue and breast hyperplasia tissue（χ~2=24.63-27.53,P〈0.05）.The expressions of BRCA1 and BRCA2 in the cancer were not associated with the patient age,tumor size,histology staging,lymph node metastasis and pathologic type（P〉0.05）. Conclusion Downregulation of BRCA1 and BRCA2 expressions may be an important factor contributing the occurence of breast cancer,but the correlation between the two expressions and clinicopathologic characteristics is not obvious.
