摘 要:目的联合检测PTEN、p14ARF、Survivin在肺癌中的表达及相互关系,为肺癌诊断、监测发展及基因治疗提供理论依据。方法用免疫组织化学法检测2009年1月~2015年12月本院87例支气管肺癌组织和13例正常肺组织中PTEN、p14ARF、Survivin基因的表达及其相关性。结果 PTEN、p14ARF表达在正常肺组织高于肺癌,而Survivin则相反;PTEN、p14ARF表达随肺癌细胞恶性程度增加而减少,而Survivin则相反,且在高、中、低分化各组间表达差异有统计学意义(P<0.01);在临床分期组随临床分期的增大,PTEN、p14ARF表达减少,而Survivin表达增强,且三者在Ⅰ期、Ⅱ期、Ⅲ期、Ⅳ期表达比较,差异有统计学意义(P<0.01)。PTEN、p14ARF有协同作用(r=0.6489,P<0.01),Survivin与PTEN,与p14ARF互为反作用(r=-0.5743,P<0.01;r=-0.6860,P<0.01)。结论 PTEN、p14ARF和Survivin共同调控肺癌细胞的凋亡和增殖,为三者在肺癌中的联合治疗提供理论依据。Objective To study the expression and the relationship of PTEN,p14^ARF and Survivin in lung carcinoma and offer more scientific theoretic basis for treatment and diagnosis of lung carcinoma.Methods 87 cases of primary bronchogenic carcinoma tissues and 13 cases normal lung tissues in our hospital from January 2009 to December 2015 were detected by SP immunohistochemical method,and their expression and correlation were studied.Results The positive expression of PTEN,p14^ARF were higher in normal lung tissue than primary bronchogenic carcinoma,the expression of Survivin was in contrast.The expression of PTEN,p14^ARF were reduced with poorly,moderately and welldifferentiated group,by contrast,the expression of Survivin was increasing(P〈0.01).The expression of PTEN,p14 ARFwere reduced,while the expression of Survivin was increasing with clinical stages enlarging in clinical stages group,PTEN,p14^ARF and Survivin among stageⅠ,Ⅱ,Ⅲ,Ⅳ had statistical difference(P〈0.01).Among PTEN,p14^ARF and Survivin were correlatively analyzed in lung carcinoma.There is a positive correlation between PTEN and p14ARF(r=0.6489,P〈0.01),with negative correlation between Survivin and PTEN(r=-0.5743,P〈0.01)or Survivin and p14ARF(r=-0.6860,P〈0.01).The three gene are associated with the primary bronchogenic carcinoma.Conclusion PTEN,p14^ARF,Survivin play a combined role in the apoptosis and proliferation of pri mary bronchogenic carcinoma.It is important to improve our treatment and diagnosis of primary bronchogenic carcinoma.
