摘 要:目的研究γ-氨基丁酸受体(GABAA-R)激动剂蝇蕈醇(muscimol)预处理是否能有效拮抗Aβ(25-35)所致的神经毒性,并探讨神经保护作用的机制。方法采用含有Neurobasal及B-27的无血清细胞培养技术体外培养新生SD大鼠的海马神经元,并用神经元特异性烯醇化酶(NSE)免疫细胞化学染色;采用MTT法测细胞活力,摸索药物作用细胞的合适浓度和时间;通过Western blot方法检测各组细胞GABAA-R蛋白α1亚基的表达水平。结果 muscimol浓度为1.0μmol/L组的海马神经元细胞活力最高。Western blot检测发现在加入muscimol 4天时GABAA-R蛋白α1亚基的表达明显增加。结论GABA-R激动剂muscimol可直接作用于海马神经元细胞而发挥神经保护作用。Objective To study the receptor of gamma amino butyric acid (GABAA-R) agonist muscimol pretreatment whether could effectively antagonize Aβ-induced neurotoxicity, and to explore the mechanism of the neuroprotective effect. Methods The serum-free medium containing Neurolbasal and B-27 was used to culture hippocamol neurons isolated from newborn SD rats. And technology of neuron-specific enolase (NSE) immunocytochemistry was used to identify the neurons. MTT method was used to measure neurons cell viability for researching the suitable concentration and time of the muscimol. Western blot was used to detect the protein of GABAA-Rα1 after extracting total protein from the neurons. Results The group of concentration of 1.0 mmol/L has the highest cell viability in all groups. The expression of GABAA-Rα1 was obviously increased by muscimol exposure for 4 days. Conclusion Muscimol can be directly applied to the hippocampal neurons and exert neuroprotection.
