摘  要:目的:探讨不同剂量浓缩牛肉汤对局灶性脑缺血大鼠脾脏IL-4和TNF-α表达的影响。方法:将60只雄性SD大鼠随机分入组I(假手术组I)、组II(假手术组II)、组III(模型组)、组IV[低剂量浓缩牛肉汤组200 mg/(kg·d)]、组V[中剂量浓缩牛肉汤组400 mg/(kg·d)]和组VI[高剂量浓缩牛肉汤组600 mg/(kg·d)],每组各10只。组IV~组VI大鼠在造模后分别以200、400、600 mg/(kg·d)浓缩牛肉汤灌胃,组II予600mg/(kg·d)浓缩牛肉汤灌胃,组I和组III以等量生理盐水灌胃。于术后2 h和7 d分别进行大鼠神经功能缺损评分,于术后7 d测量脾质量指数和脑梗死体积,并用酶联免疫吸附测定(ELISA)法检测脾脏TNF-α和IL-4水平。结果:术后2 h和7 d,与组III相比,组V和组VI大鼠神经功能缺损评分下降有统计学意义(P〈0.05);术后7 d,组I和组II大鼠脾指数、脾脏IL-4和TNF-α表达无差异;与组III相比,组V和组VI大鼠脾指数上升,脑梗死体积减少,TNF-α表达水平降低,组VI大鼠脾脏IL-4表达显著上升(P〈0.05)。结论:浓缩牛肉汤可改善大鼠神经功能缺损,减少脑梗死体积,升高脑缺血大鼠脾指数,降低脾脏TNF-α水平,升高脾脏IL-4表达,且呈剂量依赖性。Objective:To discuss the effect of concentrated beef soup(CBS) on expressions of spleen IL -4 and TNF - α in rats with focal cerebral ischemic injury. Methods:Sixty male SD rats were randomly divided into group I (sham I group), group II (sham II group), group III( model group), group IV (low dose CBS group), group V (middle dose CBS group) and group VI (high dose CBS group) ,with 10 rats in each group. After modeling,the rats in group IV,V and VI were lavaged with CBS at the doses of 200,400,600 mg/( kg · d) espectively. The rats in group II were lavaged with CBS of 600 mg/( kg ·d) and those in group I and group III was lavaged with the same volume of normal saline. At 2 h and 7 d after operation,the neurological impairment was assessed with Longa scale. On 7 d after operation,the spleen index and infarct volume fractions were assessed. The expressions of spleen IL - 4 and TNF -α were also measured on 7 d by using enzyme -linked immuno sorbent assay. Results:On 2 h and 7 d after operation, comparing with group III, the neurological impairment score' s decline of rats in group V and group VI was statistically significant (P 〈 0.05). On 7 d after operation, comparing with group III, the spleen index increased, the infarct volume infarction and the level of TNF - α decreased in group V and group VI ( P 〈 0.05 ) and the levels of IL - 4 increased in group VI ( P 〈 0.05 ). Conclusions:CBS could improve the neurological function of rats with cerebral ischemia, reduce the cerebral infarction volume, increase the spleen index, decrease the level of spleen TNF - α and increase level of spleen IL - 4, in a dose - dependent manner.
