摘  要:为了获得生防放线菌高产菌株,以生防链霉菌H3为研究对象,在单因素紫外线、微波、Na NO_2浓度诱变的基础上,采用响应面法分析优化各因素,筛选一株突变株链霉菌对豇豆腐皮镰孢根腐病菌的抑制效果较高。结果表明,出发菌株链霉菌H3对豇豆腐皮镰孢根腐病菌抑制率为34.5%。单因素试验筛选确定最佳诱变的紫外光照时间为20 s,微波加热时间为10 s,Na NO_2浓度为0.2 mol/L。响应面法分析优化各因素,并进行方差分析确定最佳工艺条件是紫外光照时间为18.27 s,微波加热时间为9.26 s,Na NO_2浓度为0.20 mol/L。在此条件下获得了一株突变株链霉菌H3-18对豇豆腐皮镰孢根腐病菌抑制率为79.75%,比出发菌株链霉菌H3对豇豆腐皮镰孢根腐病菌的抑制率提高了56.74%,比实测值(78.85%)提高了1.57%,证实了该模型的有效性。将突变株生防链霉菌H3-18,连续5次传代遗传性能较稳定。In order to obtain a high yield strain of biocontrol actinomyce strains, an antibiotic Streptomyces H3 was used as the research object. Based on single factor of mutation(ultraviolet, microwave, concentration of Na NO2), a mutant strain of Streptomyces was screened on Fusarium solaniof high inhibitory effect by mutation breeding of response surface method. Results show that original strain of Streptomyces H3 on inhibition rate of Fusarium solaniwas 34.5%. In the single factors experiment, the optimum mutation conditions were selected that UV light time was 20 s, microwave heating time was 10 s, concentration of Na NO2 was 0.2 mol/L. With response surface method the analysis and optimization of various factors, the optimum mutation conditions which analysis of variance was confirmed were UV light time for 18.27 s, microwave heating time for 9.26 s, concentration of Na NO2 for 0.2 mol/L. A mutant strain of Streptomyces H3-18 on inhibition rate of Fusarium solaniwas 79.75%. The inhibition rate of Streptomyces H3-18 was increased by 56.74% than that of original strain Streptomyces H3, and increased1.57% than the measured value(78.85%). The validity of the model was confirmed. The genetic property of mutant strain of Streptomyces H3-18 were stable, the 5 successive generations of which was inherited.
