摘  要:为获得高产DHA的优良菌株,利用低能离子束生物技术对裂壶藻(Schizochytrium limacinum)ATCC20888进行诱变选育。以平板、试管培养、显微观察为初筛指标;以生物量、油脂产量、含油率、DHA产量为综合复筛指标。最终选育出一株高产突变株,命名为15k-160s-2-3,菌株生物量为47.66 g/L,油脂产量为14.42 g/L,DHA产量为5.48 g/L,相比于出发菌株DHA产量提高了近35%,经连续10代发酵培养,其遗传性能稳定,藻油脂肪酸组成简单,利于提纯,具有良好的商业前景。In order to obtain high-yielding good strains of DHA, this paper applied the ion beam biotechnique of low energy to the mutagenesis and breeding of Schizochytrium limacinum ATCC20888. With the growth of flat plate, tube culture and microscopic observation of Sudan black B stain as the primary screening indexes, and the biomass, yield of oil, oil content and yield of DHA as composite indicators of the second screening, a high-yield mutant strain was finally selected, which was named 15k-160s-2-3. Its biomass was 47.66 g/L, the yield of oil was 14.58 g/L, and the yield of DHA was 5.81 g/L, which has been increased by nearly 35% compared with the original strain. After fermentation cultivation for 10 generations, its heredity was stable and fatty acid composition of algae oil was simple, which was conducive to purification with good commercial prospects.
