摘 要:目的:研究microRNA-100(miR-100)对膀胱肿瘤细胞株T24细胞增殖、迁移和侵袭的影响。方法:用miR-100模拟物转染T24细胞株,构建高表达miR-100细胞株;CCK-8方法检测细胞增殖能力;小室迁移实验和小室侵袭实验检测细胞迁移能力和侵袭能力。结果:模拟物转染后,miR-100表达在T24细胞中提高23.4倍,差异有统计学意义(P<0.05);CCK-8细胞增殖实验提示T24细胞实验组增殖能力弱于对照组,差异有统计学意义(P<0.05);过表达miR-100后,T24细胞迁移能力和侵袭能力均下降(P<0.05)。结论:过表达miR-100能抑制膀胱肿瘤细胞株T24的增殖、迁移和侵袭。Objective: To evaluate the effects of microRNA- 100( miRNA- 100) on cell proliferation,migration and invasion of bladder cancer cell line T24. Methods: Using miR- 100 mimics to transfect T24 cells to establish miR- 100 cell strain. Cell proliferation was assessed by cell counting assay( CCK- 8). Transwell migration assay and traswell invasion assay were performed to predict cell migration ability and invasion ability. Results: The expression of miR- 100 after transfected by miR- 100 was 23. 4 folds in T24 cells,the differences were statistically significant( P〈 0. 05). CCK- 8 cell proliferation experiments suggested that T24 cells in experimental group were less than the control group,the difference was statistically significant( P〈 0. 05). The overexpression of miR- 100 significantly downregulated cell migration and cell invasion of bladder cancer cell line T24( P〈 0. 05). Conclusion: The overexpression of microRNA- 100 inhibits cell proliferation,migration and invasion in bladder cancer cell line T24.
