摘 要:目的探讨斑贞1号、川芎嗪(TMP)和五氟尿嘧啶(5-FU)联合逆转人胃癌细胞系SGC-7901/ADR与ERCC1表达的影响。方法采用半定量逆转录聚合酶链反应(RT-PCR),监测人胃癌细胞SGC-7901/ADR细胞在5-FU组、斑贞1号组、斑贞1号+5-FU组、斑贞1号+5-FU+TMP组切除修复交叉互补基因1(ERCC1)m RNA的表达情况。结果 5-FU组与阴性对照组比较,ERCC1 m RNA表达差异无统计学意义(P>0.05),而5-FU联合中药斑贞1号及TMP处理组ERCC1m RNA表达量明显升高(P<0.05)。结论斑贞1号、TMP和5-FU联合逆转SGC-7901/ADR细胞耐药性可能与ERCC1基因相关,为当前胃癌的治疗提供了新的理论依据。Objective To explore the effects of combined Banzhen 1, TMP and 5-FU on reversing human gastric cancer cell line SGC-7901/ADR and the expression of ERCC1. Methods Semi-quantitative RT-PCR was applied to monitor the expression of ERCC1 mRNA in human gastric cancer cell SGC-7901/ADR cell in 5-FU group; Banzhen 1 group; Banzhen 1 group+5-FU group; Banzhen 1+5-FU+TMP group. Results 5-FU group and negative control group were compared, and the difference of ERCC1 mRNA expression was not significant (P〉0.05), but ERCC1 mRNA ex pression volume in 5-FU combined with TCM Banzhen 1 group and TMP group was significantly improved (P〈0.05). Conclusion Combined Banzhen 1, TMP and 5-FU in reversing drug resistance of SGC-7901/ADR cell may be correlated with ERCCI gene, which provides new theoretical evidence for the treatment of gastric cancer.
