摘 要:采用ITS序列分析方法,对供试18株(4株野生菌株、7株工厂化栽培菌株、5株经钴60辐照诱变菌株和2株搭载神十的航天诱变菌株)茶树菇菌株进行研究。结果表明,供试菌种的ITS序列长度为703~721 bp,与Gen Bank数据库中茶树菇菌种的ITS序列相似度为99%以上,在种的水平上证明供试菌种为茶树菇。运用构建的系统发育树将供试菌种聚为6个类群,其中Cha3与Cha3shen,AS-2与AS-2-600,AS-1与AS-1shen、AS-1-700,分别聚在了不同的类群,说明这7株茶树菇菌株可能由于辐照或航天诱变后使得ITS序列存在着种内的变异。ITS序列分析结果从系统发育角度反映出了研究菌株的遗传关系,是进行茶树菇菌株遗传分析及科学鉴定的重要工具。18 tested Agrocybe aegerita strains( 4 wild strains,7 factory cultivation strains,5 Cobalt-60 irradiation induced strains,2 aerospace mutation strains carried by the Shenzhou 10 spacecraft) by ITS sequence analysis were studied. The results showed that the lengths of ITS sequences of Agrocybe aegerita strains were 703- 721 bp,and their ITS sequence similarity in the Gen Bank database was more than 99%,which indicated that the tested strains were Agrocybe aegerita at the specific level. Phylogenetic tree was also constructed that the tested strains were clustered into six groups,of which Cha3 and Cha3 shen,AS-2 and AS-2-600,AS-1 and AS-1shen and AS-1-700 were respectively clustered into different groups,which indicated that there were intraspecific variations for ITS sequences of these seven strains under the conditions of irradiation or aerospace mutation. The phylogenetic relationships were distinctly delineated using ITS sequencing,which was an important tool in the genetic analysis and identification of Agrocybe aegerita.
