摘 要:目的探讨2014年广州4株1型登革病毒分离株基因型及来源。方法 4株2014年广州市1型登革病毒分离株提取RNA,逆转录PCR扩增包膜蛋白基因并测序,对其进行系统进化分析,结合流行病学资料进行生物信息学分析。结果 4株2014年1型登革病毒分离株分别来自不同的基因亚型,即基因I型(1株)和基因V型(3株),同广州或广东其他地区2013年分离株高度相似,最初由东南亚或印度输入我国。结论广州市2014年1型登革病毒为由商贸旅游由外地输入到广东地区,并已逐渐本地化,须进一步从蚊媒中证实。Objective The genetic relationships among four dengue virus serotype 1(DENV-1) isolates from the Guangzhou dengue epidemic of 2014 were studied by sequence analysis of the envelope(E) genes. Methods The whole length of E gene from four DENV-1 isolates was amplified by reverse transcriptase-polymerase chain reaction(RT-PCR)and then sequenced.The phylogenetic tree was drawn by neighbor-joining method.The bioinformatics analysis was performed by combining the phylogenetic information and the epidemiology data. Results Among 4 isolates revealed an epidemic strain in the outbreak.Phylogenetic study demonstrated that the 2014 Guangzhou isolates were of genotype I and genotype V and similar from Guangdong DENV-isolates from 2013,which were initially imported from Southeast Asia and India. Conclusion Our result showed the same genotype in Guangdong isolates of 2013 and 2014 suggested the 2014 Guangzhou DENV-1 isolated strains were imported from nearby countries by trade and tourism and gradually autochthonous; further prove of their origins was needed.
