宫颈癌患者外周血和宫颈组织中髓源性抑制细胞的检测及意义

摘　　要：目的:检测髓源性抑制细胞(myeloid-derived suppressor cells,MDSC)在宫颈癌患者外周血和癌组织中的比例及亚群分布,初步探讨其临床意义。方法:采用流式细胞术分别检测17例宫颈癌患者治疗前和20例健康者外周血、17例宫颈癌组织和15例正常宫颈组织细胞悬液中MDSC比例及其亚群分布,并分析其与鳞状细胞癌抗原的相关性。结果:与健康者相比,宫颈癌患者外周血中MDSC比例明显增高(P<0.01),以CD15+粒细胞样亚群为主。与正常宫颈组织细胞悬液相比,癌组织细胞悬液CD45+细胞的比例明显增加(P<0.05),但细胞中未见CD33+表面标志的表达。宫颈癌患者外周血MDSC比例与鳞状细胞癌抗原值呈正相关(r=0.715 4,P<0.05)。结论:外周血中粒细胞样的MDSC可能与宫颈癌的发生发展密切相关。Objective： To investigate the frequency and clinical significance of myeloid-derived suppres- sor cells （MDSC） in peripheral blood and tumor loci of the patients with cervical cancer. Methods： Flow cytometry was used to evaluate,the proportion of MDSC in the blood of 17 patients with cervical cancer pre- therapy and 20 healthy people. The proportions of MDSC in the tissue cell suspension were evaluated in 17 patients with cervical cancer and 15 healthy cervical tissues. Results： Compared with healthy volunteers, the proportions of circulating MDSC from cervical cancer were significantly higher（P 〈 0.01 ） ; and the MD- SC in peripheral blood mainly consists of CD15 ＋ granulocyte subgroup. The proportions of tissue cell sus- pension CD45＋ cell from cervical cancer were significantly higher, compared with normal tissue cell suspen- sion （ P 〈 0.05 ） ; but in tissue cell suspension CD33 ＋ cell was not found. The proportion of MDSC was posi- tively correlated with the frequency of value of squamous cell carcinoma antigen in peripheral blood of pa- tients with cervical cancer （r = 0. 715 4, P 〈 0.05）. Conclusion ： Granulocyte subgroup MDSC in the pe- ripheral blood of patients with cervical cancer may be associated with the development of cervical cancer.
