摘  要:建立了早胜牛心肌中牛磺酸的高效液相检测方法并优化其提取工艺。以异硫氰酸苯酯为衍生剂,HPLC法检测牛磺酸浓度。采用Shim-pack VP-ODS C18色谱柱,流动相流速1.0 m L/min,检测波长254 nm,柱温36℃。以早胜牛心肌为原料,研究料液比、提取时间、水解时间、提取温度对牛磺酸提取率的影响,通过响应面分析法优化其最佳提取工艺。牛磺酸线性范围为0.37~90μg/m L,相对标准偏差(RSD)在1.5%~2.3%(n=4)之间。当料液比1∶5、提取时间55 min、水解时间20.5 h、提取温度85℃时,牛磺酸最高提取率可达3.76 mg/g。方法对于早胜牛中牛磺酸提取和检测应用具有一定参考价值。A method to optimize and determine taurine in myocardial of zaosheng cattle by high performance liquid chromatography( HPLC) was described. After pre-column derivatization with othiocyanate,the content of taurine was determined by HPLC. The chromatographic condition as follows. Shim-pack VP-ODS C18 column was adopted,the rate of mobile phase was 1. 0 m L/min,the detection wavelength was set at 254 nm and the column temperature was 36° C. The effects of ratio of solvent to material,treatment time,hydrolysis time,extraction temperature on the yield of taurine were investigated by response surface methodology,which was detected by high performance liquid chromatography( HPLC) with pre-column derivatization. The method showed a good linearity within the concentration range of 0. 37 ~ 90 μg/m L,RSD of 1. 5% ~ 2. 3%( n = 4). The optimum extraction conditions of taurine were got that were ratio of material to solvent 1: 5,treatment time 55 min,hydrolysis time 20. 5 h and extraction temperature 85° C. Under these conditions,the yield of taurine was3. 76 mg/g. The higher rate of extraction and detection of taurine from zaosheng cattle for industrial production has a certain reference value.
