紫杉醇脂质体联合中药姜黄素对小鼠转移性肺癌的影响

摘　　要：目的:观察紫杉醇脂质体联合中药姜黄素腹腔注射或雾化吸入对小鼠转移性肺癌的影响。方法:小鼠于皮下接种瘤细胞悬液复制肿瘤模型。造模后第7、10、13、16、19、22 d,模型组雾化吸入生理盐水;紫杉醇腹腔注射组及雾化组分别腹腔注射或雾化吸入5 mg·kg-1紫杉醇脂质体;紫杉醇腹腔注射+姜黄素灌胃组及紫杉醇雾化+姜黄素雾化吸入组腹腔注射及雾化吸入5 mg·kg-1紫杉醇脂质体,隔天灌胃及雾化吸入50 mg·kg-1姜黄素溶液;环磷酰胺组腹腔注射60 mg·kg-1环磷酰胺;5d为1个周期,干预22 d。造模后第30 d处死小鼠,剖取完整瘤块,计算抑瘤率。结果:联合用药组大鼠瘤重明显低于单用紫杉醇组(P<0.05),且抑瘤率均高于单用紫杉醇组的抑瘤率;但紫杉醇腹腔注射联合姜黄素灌胃组小鼠死亡较多,雾化吸入组小鼠未见死亡。结论:紫杉醇联合姜黄素雾化吸入具有较好的抑制肿瘤作用,且可以维持肿瘤小鼠治疗期间存活率,具有抗肿瘤治疗的潜力。Objective： to observe the effect of Liposomal paclitaxel and Curcumin on spontaneous metastatic lung cancer of mice through intraperitoneal injection or aerosol inhalation. Methods： Mice were received subcutaneous inoculation of tumor cells to form tumor model. After inoculation 7,10,13,16,19,22 days,model group mice were given aerosol inhalation with normal saline,paclitaxel group received Liposomal paclitaxel 5 mg ·kg- 1by intraperitoneal injection or aerosol inhalation,paclitaxel combining with curcumin group received Liposomal paclitaxel 5 mg·kg- 1,the first day and curcumin the next day 50 mg·kg- 1by intregastric adminis tration or aerosol inhalation,Cyclophosphamide group was treated by cyclophosphamide 60 mg·kg- 1through intraperitoneal injection,5 days for a cycle intervention in 22 days. on the 30 th day after inoculation,mice were sacrificed,moved complete tumor tissue and calculated the anti- tumor rate. Results： Combination drug therapy could decrease tumor weight significantly（ P〈0. 05）,and the anti- tumor rate was higher than that of single medication group,however,the way of intraperitoneal injection made more mice die than aerosol inhalation.Conclusion： Liposomal paclitaxel combining with Curcumin could inhibit tumor growth by aerosol inhalation,and keep the tumor mice survival during treatment. The research suggested that Liposomal paclitaxel and Curcumin combination has the potential of cancer therapy.
