宫颈癌组织中miR-218和Survivin的表达及相关性研究

摘　　要：目的探讨微小RNA-218(miR-218)和凋亡抑制基因Survivin在宫颈癌中的表达情况及临床意义。方法采用实时荧光定量PCR(RT-PCR)检测48例宫颈癌及48例正常宫颈组织中miR-218和Survivin基因的表达,免疫组化Envision法检测相应癌组织中Survivin蛋白的表达,分析miR-218和Survivin表达的相关性及其与患者临床病理特征的关系。结果 miR-218在宫颈癌组织中的表达明显低于正常组织,差异有统计学意义(P<0.05);其表达与癌细胞分化程度及淋巴结转移有关(P<0.05)。Survivin在宫颈癌组织中的表达率为85.42%(41/48),显著高于正常宫颈组织,差异有统计学意义(P<0.05);Survivin基因的表达与癌组织分化程度具有相关性(P<0.05)。MiR-218和Survivin基因的表达呈负相关(P<0.05)。结论 MiR-218在宫颈癌中的表达率降低,可能与宫颈癌的增殖、侵袭转移有关。Objective To investigate the expression and clinical significance of microRNA-218（ miR-218） and apoptosis inhibitor gene Survivin in cervical cancer. Methods By using quantitiative PCR,the expression of miR-218 and Survivin gene was detected in 48 cases of cervical carcinoma and 48 cases of normal cervical tissues; to detect the expression of Survivin protein in corresponding cancer tissues by immunohistochemical Envision method; the correlation between the miR-218 and Survivin expression and its relationship with the clinical and pathologic characteristics were analyzed. Results The expression of miR-218 in cervical cancer tissues was significantly lower than that in normal tissues,with the difference statistically significant（ P 〈 0. 05）; and it was also related with tissue differentiation degree and lymph node metastasis（ P 〈 0. 05）. The expression rate of Survivin in cervical cancer tissues was 85. 42%（ 41 /48） and significantly higher than the normal tissues,with the difference statistically significant（ P 〈 0. 05）; the expression of Survivin was also related with tissue differentiation degree（ P 〈0. 05）. The expression of miR-218 and Survivin showed a negative correlation（ P 〈 0. 05）. Conclusion The expression rate of miR-218 in cervical carcinoma is low,which may be related to the proliferation,invasion and metastasis of cervical cancer.
