摘　　要：目的探索p53凋亡刺激蛋白抑制因子(iASPP)在结直肠癌组织/细胞中的表达及iASPP沉默后对结直肠癌细胞增殖的影响。方法该研究首先检测临床样本结直肠癌组织和癌旁正常组织中iASPP mRNA的表达差异、结直肠癌细胞系和正常结肠上皮细胞系(HCoEpiC)mRNA和蛋白质iASPP表达差异;然后在人结肠癌细胞SW480中转染iASPPsiRNA,用实时荧光定量聚合酶链反应(RT-qPCR)检测iASPP mRNA表达效率,用免疫印迹试验(Western blot)检测iASPP的蛋白表达水平;最后用MTT法和Brd U法检测沉默iASPP对SW480细胞增殖的影响。结果结直肠癌组织中,iASPP mRNA表达相比癌旁正常组织上调;结直肠癌细胞LoVo、SW620和SW480中iASPP mRNA和蛋白质表达相比正常结肠上皮细胞系(HCoEpiC)均上调;在SW480细胞中转染iASPP-siRNA可下调iASPP的表达;沉默iASPP可抑制结直肠癌细胞的增殖。结论沉默iASPP基因抑制结直肠癌细胞的增殖。【Objective】To explore the expression of inhibitory member of apoptosis-stimulating protein of p53 family（iASPP） in colorectal cancer tissue samples and cell lines, and the effect of knockdown of iASPP on the proliferation of colorectal cancer cells. 【Methods】iASPP was selected as the candidate target gene. Firstly, the expression levels of iASPP mRNA and protein in colorectal cancer tissues and cells and adjacent normal tissues and cells were detected. Then iASPP-siRNA was transfected into SW480 colorectal cancer cells, the expression efficiency of iASPP mRNA was detected by real-time quantitative reverse transcription polymerase chain reaction（RT-qPCR）, and the iASPP protein expression was detected by Western blot. Finally the effect of downregulation of iASPP by iASPP-siRNA on the proliferation of SW480 cells was detected by MTT and Brd U assays.【Results】The expression of iASPP mRNA was increased in colorectal cancer tissue samples as compared with adjacent normal tissue; at the same time, the expression of iASPP mRNA and protein was increased in colorectal cancer cell lines（LoVo, SW620 and SW480） compared with colonic epithelial cell line（HCoEpiC）; iASPP-siRNA down-regulated the expression of iASPP, and inhibits the proliferation of SW480 cells.【Conclusion】Knockdown of iASPP inhibits the proliferation of colorectal cancer cells.
