摘  要:应用MicroRNA(miRNA)芯片技术筛选患者化疗前后血浆中表达差异明显的miRNA,发现与化疗敏感性相关的miRNA,为临床化疗药物敏感性预测提供新的方法.共收集28例非小细胞肺癌患者含铂双药化疗前后的血液标本,通过miRNA基因芯片进行差异表达的miRNA筛选,并采用实时定量RT-PCR方法对部分差异表达的miRNA进行可靠性验证.对于化疗有效的非小细胞肺癌患者,筛选出4个明显表达上调的miRNAs,分别是:Hsa-miR-502-3p,Hsa-miR-26a-5p,Hsa-miR-210,Hsa-miR-191-5p;筛选出3个明显表达下调的miRNAs:Hsa-miR-141-3p,Hsa-miR-193b-3p,Hsa-miR-122-5p.本研究证明Hsa-miR-502-3p,HsamiR-26a-5p,Hsa-miR-210,Hsa-miR-191-5p的表达上调及miRNA Hsa-miR-141-3p,Hsa-miR-193b-3p,Hsa-miR-122-5p的表达下调与非小细胞肺癌化疗药物敏感性相关.MicroRNA( miRNA) array is adopted to screen the differential expression of miRNA in non- small cell lung cancer( NSCLC) and to identify the miRNAs related to chemosensitivity,which is a new way to predict the effect of medicine. Plasma is extracted from 28 patients before and after double drugs chemotherapy containing cisplatin. Identification of differentially expressed MicroRNAs is carried out by miRNA array. Real time quantitative PCR is applied to verify the reliability of miRNA array results. In chemotherapy sensitive patients,4up- regulated miRNAs are Hsa- miR- 502- 3p,Hsa- miR- 26a- 5p,Hsa- miR- 210 and Hsa- miR- 191- 5p; and 3 down- regulated miRNAs are Hsa- miR- 141- 3p,Hsa- miR- 193b- 3p and Hsa- miR- 122- 5p. It is concluded that the differential expression of miRNA is related to chemosensitivity.
