摘  要:目的探讨益气复生方对小鼠胃癌脾气虚证的代谢组学影响。方法 20只健康雄性小鼠,140 d白醋+2-乙基亚硝胺干预条件下建立胃癌脾气虚证小鼠模型,取存活的15只小鼠中表型和体质量相近的12只,随机分为对照组和实验组,每组6只。实验组小鼠给予40 g/kg体质量益气复生方灌胃,对照组给予相同质量的蒸馏水,均连续14 d处理。经变量重要性投影(VIP)的参数评价可能的小鼠血清生物标志物,对两组小鼠血清生物标志物进行核磁共振(1H-NMR)检测,观察两组小鼠相关代谢产物的变化。结果实验组小鼠经过益气复生方的治疗后体质量、摄食量、活动度增加,大便部分成型,毛发由枯槁恢复为正常黑棕色,有光泽。对照组小鼠与建模成功时无明显变化。经变量重要性投影(VIP)的参数评价可能的胃癌脾气虚证小鼠血清生物标志物包括3-羟基丁酸、低密度脂蛋白/极低密度脂蛋白、甲硫氨酸、谷氨酸、缬氨酸、谷氨酰胺、葡萄糖、甘油三酯、不饱和脂肪酸和胆碱。实验组小鼠血清1H-NMR谱中代谢产物中3-羟基丁酸、甲硫氨酸、缬氨酸、谷氨酰胺的含量低于对照组(P<0.05),低密度脂蛋白/极低密度脂蛋白、谷氨酸、甘油三酯、不饱和脂肪酸和胆碱的含量则高于对照组(P<0.05),葡萄糖的含量与对照组比较差异无统计学意义(P>0.05)。结论益气复生方对小鼠的胃癌脾气虚证具有显著的治疗效果,其作用可能是通过调控能量代谢水平及脂质代谢水平的来实现的。Objective To explore the metabonomic effects of Yiqifusheng recipe on spleen-qi deficiency syndrome of mice with gastric cancer. Methods 20 healthy male mice were used to establish the model of spleen-qi deficiency syndrome and gastric cancer processed with white vinegar and 2-ethyl-N-nitrosamines for 140 days. 12 mice with similar phenotype and body mass were selected from survival groups( 15 mice),then randomly divided into control and experimental groups,six mice per group. The mice in experimental group were given 40 g / kg Yiqifusheng recipe,and mice in the control group were received the same quantity of distilled water for 14 days. Possible serum biomarkers were evaluated by variable importance projection( VIP) parameters,and the serum biomarkers of mice were detected by hydrogen proton nuclear magnetic resonance spectroscopy(1H-NMR) in two mice groups,and the changes of metabolites related to the serum biomarkers were observed. Results The mice in the experimental group were treated with Yiqi Huansheng Recipe. The body weight,food intake and activity of the mice were increased,the stool was partially formed and the hair was returned from normal to dark brown. While,there was nosignificant changes in the mince of control group. The serum biomarkers of mice with spleen-qi deficiency syndrome and gastric cancer mice evaluated by VIP were 3-hydroxybutyric acid,LDL / VLDL,methionine,glutamic acid,valine,glutamine,glucose,triglycerides,unsaturated fatty acids and choline.1H-NMR spectra of mice serum metabolites showed that: the content of 3-hydroxybutyric acid,methionine,valine,glutamine in the experimental group were lower than those in the control group( P〈 0. 05),however,the content of LDL / VLDL,glutamic acid,triglycerides,unsaturated fatty acids and choline were higher in the experimental group than those in the control group( P〈 0. 05). The content of glucose had no significant effect on mice serum metabolites( P〉 0. 05) in the 1H-NMR spectrum. Conclusion Yiqifusheng have a
