心脑宁片对大鼠脑缺血模型凋亡基因Bcl-2和Bax蛋白表达的影响

摘　　要：目的 研究心脑宁片对大鼠脑缺血模型脑组织匀浆中凋亡基因Bcl-2、Bax蛋白表达的影响。方法取体重180~200 g雄性Wistar大鼠36只,随机分为3组：第一组为假手术组,其余2组造脑缺血模型。模型Ⅰ、Ⅱ组分别灌服0.5%羧甲基纤维素钠（CMC）和同体积的0.75 g/kg含量的心脑宁片混悬液,采取酶联免疫技术测定匀浆中Bcl-2及Bax的含量。结果 与假手术组比较,模型Ⅰ组大鼠脑组织匀浆中Bcl-2含量显著增加（P〈0.05）,而Bax含量非常显著增加（P〈0.01）;与模型Ⅰ组比较,模型Ⅱ组有显著高的Bcl-2含量（P〈0.05）和显著低的Bax含量（P〈0.05）,且Bcl-2/Bax显著升高（P〈0.05）。结论 心脑宁片可以提高抑凋亡基因含量,降低促凋亡基因含量,明显增加抑凋亡基因与促凋亡基因的浓度比率,表明其对脑组织有着明显的保护作用。Objective To study the effect of Xinnaoning tablets on the Bcl-2,Bax levels of the rat cerebral ischemia model. Methods Thirty-six male Wistar rats were divided into three groups. The first group was the sham operation group,and the other two groups were made into the cerebral ischemia model,i.e model group Ⅰ was fed with CMC suspension and model group Ⅱ was fed with 0.75 g/kg suspension of this tablets,and then the Bcl-2,Bax levels in three groups were detected by ELASA. Results Compared with the sham operation group,model group I had the higher Bcl-2 level（P〈0.05）,and significantly higher Bax level （P〈0.01）;Compared with the model group Ⅰ ,model group Ⅱ had significantly higher Bcl-2 level and Bel-2/Bax （P〈0.05） ,but lower Bax level （P〈0.05）. Conclusion Xinnaoning tablets could have a protective effect on the brain tissue,which showed by it uplifting the level of suppression-apoptosis gene,decreasing promotion-apoptosis gene,and significantly in creasing the ration of both.
