摘 要:目的:对比含10%、20%、40%胎牛血清(fatal bovine serun,FBS)的DMEM培养基对原代培养、分离纯化人牙周膜干细胞(human periodontal ligament stem cells,h PDLSCs)生物学特性的影响,为h PDLSCs的相关体内、体外研究提供理论和实验依据。方法:分别用3组含不同浓度FBS的DMEM培养基对h PDLSCs进行原代培养、分离纯化,在倒置显微镜下观察细胞形态、生长特征,用波形蛋白的免疫细胞化学染色方法进行细胞表型鉴定,MTT比色法检测细胞的增殖能力。结果:各组DMEM培养基原代培养分离的h PDLSCs形态相似,均显示正常的细胞形态和生长特征;其中含20%FBS培养基组细胞增殖力更强;3种培养条件培养的h PDLSCs表型相似,均阳性表达波形蛋白。结论:含不同浓度FBS的DMEM培养基均能在体外培养扩增h PDLSCs,并维持其干细胞特性。采用含20%FBS培养条件培养h PDLSCs可能更适合其在细胞治疗和实验研究中应用。Objective: To compare the influences of fetal bovine serum( FBS) DMEM medium( containing 10 % FBS,20 % FBS,and 40 %FBS) on biological characteristics of isolated and purified human periodontal ligament stem cells in primary culture( h PDLSCs) so as to provide theoretical and experimental basis for the research of h PDLSCs in vivo and in vitro. Methods: The primary culture and purification of h PDLSCs were performed with three groups of DMEM medium containing different concentrations of FBS,and cell morphology and growth characteristics were observed under inverted microscope. The cells were identified by immunocytochemistry of vimentin staining method,MTT colorimetric assay for cell proliferation. Result: HPDLSCs morphology was similar in the primary culture of DMEM medium with different concentrations of FBS in each group,with cell morphology and growth characteristics of normal display; the cell proliferation is stronger in the group containing 20 % FBS medium; three kinds of cultured h PDLSCs were phenotypically similar,all with positive expression of vimentin. Conclusion: DMEM medium containing different concentrations of FBS can amplify h PDLSCs in vitro,while maintaining their stem cell characteristics. The use of h PDLSCs containing 20% FBS culture conditions may be more suitable for its application in cell therapy and experimental research.
