摘 要:目的:检测非小细胞肺癌(non-small cell lung cancer,NSCLC)中肿瘤干细胞标志物乙醛脱氢酶1(aldehyde dehydrogenase1,ALDH1)和肿瘤转移抑制基因KAI1的表达情况,并分析它们在NSCLC患者中的临床病理意义。方法:采用免疫组织化学Eli VisionTMplus法检测106例NSCLC和40例正常肺组织中ALDH1和KAI1蛋白的表达情况。结果:在NSCLC组中ALDH1和KAI1蛋白的表达阳性率分别为60.4%和37.7%,对照组中ALDH1和KAI1蛋白的表达阳性率分别为10.0%和90.0%,2组表达差异均有统计学意义(P〈0.01)。ALDH1蛋白和KAI1蛋白表达阳性率在NSCLC组织的不同分化程度、淋巴结转移与否及TNM分期高低间差异均有统计学意义(P〈0.05~P〈0.01)。等级相关分析证实在NSCLC组织中ALDH1蛋白与KAI1蛋白表达呈负相关关系(P〈0.05)。结论:异常表达的ALDH1和KAI1可能参与了NSCLC的发展、浸润和转移等过程;在NSCLC患者中联合检测ALDH1和KAI1蛋白的表达有益于早期预测其浸润和转移。Objective:To explore the expressions of aldehyde dehydrogenase 1 ( ALDH1 ) of the marker of tumor stem cells and KAI1 of tumor metastasis suppressor gene in non-small cell lung cancer( NSCLC), and analyze its clinicopathological significance. Methods: The protein expressions of ALDH1 and KAI1 in 106 NSCLC specimens and 40 normal lung tissues were detected using immunohistochemical EliVisionTM plus. Results: The positive rates of ALDH1 and KAI1 protein in NSCLC tissues and control tissues were 60.4% & 37.7 % and 10.0% & 90.0% , respectively, there was significant difference between the two groups ( P 〈 0.01 ). The differences of the positive expression rates of ALDH1 and KAI1 protein in different differentiation degrees,lymph node metastasis and TNM stage in NSCLC tissues were statistically significant( P 〈 0.05 to P 〈 0.01 ). Spearman analysis indicated that the expression of ALDH1 was negative correlation with the expression of KAI1 in NSCLC tissues( P 〈 0.05 ). Conclusions:The abnormal expressions of ALDH1 and KAI1 may be involved in the development, invasion and metastasis of NSCLC. The combined detection of ALDH1 and KAI1 may be of significant value in early predicting the invasion and metastasis of NSCLC.
