摘 要:[目的]从转录组的角度研究极端嗜盐古菌Natrinema sp.J7-2的盐适应性。[方法]利用Illumina Hi Seq 2000双末端测序技术对生长在不同盐浓度下(15%Na Cl、25%Na Cl、30%Na Cl)的Natrinema sp.J7-2的转录组测序分析,并对差异表达基因进行了GO与KEGG富集。[结果]转录组测序结果表明,培养在3种不同盐浓度下的Natrinema sp.J7-2差异表达基因主要富集到氨基酸代谢、卟啉与叶绿素代谢两条途径上,氨基酸代谢主要涉及谷氨酸、天冬氨酸、丙氨酸代谢;在3组盐培养物中,有1 320个基因存在差异,其中83个共同差异表达的基因。[结论]通过转录组分析揭示Natrinema sp.J7-2菌株主要通过调控氨基酸代谢与能量代谢来适应不同盐环境。[ Objective]To explore the salt adaptation of Natrinema sp. J7 -2 based on transcriptomics sequencing. [ Methods ] Illumina HiSeq 2000 pair - end sequencing technology was employed to analyze the Natrinema sp. J7 - 2 grown under different salt concentrations (15 % NaC1,25 % NaC1 and 30% NaC1), then the differential expression analysis was carried out, and then GO and KEGG of differentially expressed genes were enriched. [ Results ] The differentially expressed genes were mainly enriched in amino acids metablism, porphyrin and chlorophyll metabolism; amino acids metabolism mainly involved glutamate, aspartic acid and alanine. And there were 1 320 differentially expressed genes and only genes 83 commonly differentially expressed genes among cells incubated in 15% ,25% and 30% NaC1 media. [ Conclusion] Natrinema sp. J7 -2 cells regulated amino acids metabolism and energy metabolism to adapt different salinity environments.
