摘 要:[目的]建立标准的Nestin-LacZ转基因小鼠脊髓损伤动物模型及评价体系。[方法]采用NestinLac Z转基因小鼠18只,随机分为三组,实验组2个,对照组1个。2个实验组均采用自行研制的啮齿类动物脊髓损伤模型打击器,质量为10 g的打击棒,A实验组用接触直径为2.0 mm的打击棒杆头压迫,压迫时间为3 min(A组,6例);B实验组按照实验设定的高度10.0 mm实施打击(B组,6例);对照组仅行椎板切除,不打击(C组,6例)。分别于术前,术后第1、2、3、4、5、6周进行转基因小鼠BBB运动评分,取出脊髓组织后,行脊髓组织HE染色。[结果]Nestin-LacZ转基因小鼠不同的脊髓损伤模型在各个时间点上BBB评分差异有统计学意义(P<0.05),A组在脊髓损伤造模术后2周,后肢功能开始恢复,而B组至术后6周均无后肢功能恢复,对照组无功能障碍。[结论]啮齿类动物脊髓损伤模型打击器采用持续慢性压迫脊髓可成功建立Nestin-Lac Z转基因小鼠稳定、理想的急性脊髓损伤模型。[ Objective] To establish a standard Nestin -LacZ transgenic mouse model of spinal cord injury and the related evaluation system. [Method]A total of 18 Nestin -LacZ transgenic mice were randomly divided into two experimental groups and one control group. In one experimental group (group A, n = 6), meanwhile, a self - developed impacting rod ( 10 g) for es- tablishment of the spinal cord injury model in rodents was applied, and compression was performed with the tip of the impacting rod ( contact diameter 2.0 mm) for 3 rain. In the other experimental group ( group B, n = 6), meanwhile, a self - developed im- pacting rod (10 g) for establishment of the spinal cord injury model in rodents was applied, and impacting was performed at a height of 10.0 mm. The mice in the control group (group C ,n =6) underwent laminectomy alone. Before injury and at 1,2,3, 4,5 ,and 6 weeks after injury, the Basso- Beattie- Bresnahan (BBB) scores were measured; the spinal cord tissues were col- lected for histological observation and then subjected to HE staining. [ Result] BBB scores showed significant differences be- tween the Nestin - LacZ transgenic mice with different types of spinal cord injury at all time points ( P 〈 0.05). The mice in group A had functional recovery in hind limbs at 2 weeks after injury,while those in group B had no functional recovery in hind limbs throughout 6 weeks after injury;the mice in group C showed no dysfunction. [ Conclusion] Continuous compression of the spinal cord with the self - developed impactor for the establishment of the spinal cord injury model in rodents can successfully establish a stable and ideal Nestin - LacZ trans- genic mouse model of acute spinal cord injury.
