摘 要:探讨ATP生物发光法对检测食源性致病菌不同生长阶段菌落数目的适用性,分析食源性致病菌不同培养时期胞内ATP含量的差异对菌落计数影响的显著性。以肠致病性大肠埃希氏菌、金黄色葡萄球菌金黄亚种和蜡样芽胞杆菌为试验对象,通过ATP生物发光法,辅以传统平板计数法,构建3种食源性致病菌各生长时期菌落浓度与荧光强度的标准曲线,并计算相应生长阶段各致病菌胞内ATP含量。结果表明:3种食源性致病菌不同生长时期相同菌落浓度对应的荧光强度对数值差异极显著(P<0.01),相同菌落浓度3种食源性致病菌各阶段胞内ATP含量差异极显著(P<0.01)。3种食源性致病菌不同生长阶段胞内ATP含量的差异对菌落计数影响显著,在实际应用中,不可将单一生长时期所建立的标准曲线作为检测标准,应对特定样品进行多次重复测定建立相应的数学模型进行分析。The ATP bioluminescence detection technology has various advantages compared with the traditional colony counting method in plates. In this study, the applicability of ATP bioluminescence technology for detection of foodborne pathogenic bacteria in different growth stage was discussed, and the significance of impact of the differences intracellular ATP content in different culture period on the number of colony was analyzed. Strains including Escherichia coli, Bacillus cereus and Staphylococcus aureus were selected in this study, and the standard curves between colony concentration in different culture stages and fluorescence intensity of these strains were established with ATP bioluminescent and plate count methods. The ATP content in different strains during stages was calculated using the standard curves.The results showed that logarithm numbers of fluorescence intensity of same colony concentration in different culture stages of the three different strains were significantly different. The ATP content of same colony concentration of three different strains was also different.Three kinds food-borne pathogens intracellular ATP content of different growth stages had significantly effect on colony number. In practical application, we cannot regard a single growth period as the testing standard and corresponding mathematical model with specific samples should be established for the comprehensive analysis.
