结直肠癌中S100A4的表达及意义

摘　　要：目的探讨S100A4在结直肠癌组织及细胞系中的表达,并分析其与结直肠癌临床病理特征的相关性。方法采用免疫组化法检测65例结直肠癌及对应癌旁正常结直肠组织中S100A4蛋白的表达;分别采用RT-PCR法和Western blot法检测S100A4 mRNA和蛋白在7个人类结直肠癌细胞系中的表达。结果 RT-PCR及Western blot检测结果显示S100A4在多数结直肠癌细胞中均有表达。免疫组化检测结果显示S100A4阳性表达主要定位于结直肠癌细胞质内,结直肠癌组织中S100A4蛋白阳性率高于癌旁正常结直肠组织(P<0.000 1)。S100A4蛋白表达与结直肠癌患者年龄、性别有关,高龄组阳性率高于低龄组(P=0.026),男性组S100A4蛋白阳性率高于女性组(P=0.038),其表达与肿瘤部位、分化程度、淋巴结转移均无相关性(P>0.05)。结论 S100A4在多数结直肠癌细胞中均有表达,在癌组织中的表达高于癌旁组织,S100A4蛋白表达与结直肠癌患者年龄、性别有关,可能为结直肠癌患者的个性化诊断及治疗提供指导。Purpose To investigate the expression of S100A4 in colorectal carcinoma tissue and cell lines, and to analyze the correla- tion between expression of S100A4 and clinical parameters of colorectal carcinoma. Methods Immunohistochemical method was used to detect the expression of S100A4 protein in 65 cases of colorectal carcinoma and its adjacent mucosa. RT-PCR and Western blot method were used to detect the expression of S100A4 mRNA and protein in seven human colorectal carcinoma cell lines. Result RT- PCR and Western blot showed S100A4 mRNA and protein expressd in most of colorectal carcinoma cell lines. Immunohistochemical re- sults showed that the expression of S100A4 protein mainly located in the cytoplasm of colorectal carcinoma. The expression of S100A4 protein in colorectal carcinoma tissue was significantly higher than that in adjacent mucosa （P 〈 0, 001 ）. Expression of S100A4 protein was found to be associated with age and sex. Elderly group was higher than that in young group （P = 0. 026 ） , male group was higher than that in female group （P = 0. 038）. No significant association was found between the expression of S100A4 and other clinicopatho- logical parameters such as location, differentiation and lymph node metastasis （ P 〉 0. 05 ）. Conclusion S100A4 may expresse in most of colorectal carcinoma cell lines. Expression of S100A4 protein in eolorectal carcinoma tissue is significantly higher than that in its adjacent mucosa, and is associated with age and sex, which may provide guidance for the diagnosis of colorectal cancer and person- alized treatment of patients.
